摘要
目的研究莫诺苷对过氧化氢诱导PC12细胞损伤的保护作用及其机制。方法MTT法检测细胞存活率,探索过氧化氢损伤模型,筛选莫诺苷浓度。将PC12细胞分为对照组、模型组、阳性药组及不同浓度莫诺苷组,分别采用MTT及乳酸脱氢酶法检测细胞存活率,采用荧光探针法检测细胞内活性氧以及丙二醛的含量,Western blot方法检测细胞内Bcl-2蛋白表达的变化。结果过氧化氢诱导PC12细胞损伤明显,与模型组相比,莫诺苷各组PC12细胞存活率显著提升,乳酸脱氢酶漏出量显著降低;莫诺苷能够降低细胞内活性氧水平,减少丙二醛的产生,上调细胞内抗凋亡蛋白Bcl-2的表达。结论莫诺苷对过氧化氢诱导的PC12细胞具有一定的保护作用。
Objective To investigate the protective effect of morroniside against cell death induced by hydrogen peroxide and the mechanism.Methods MTT assay was used to screen the concentrations of hydrogen peroxide and morroniside.PC12 cells were divided into control group,model group,positive drug group and morroniside groups of different doses.Cell viability and mortality were detected with MTT and lactate dehydrogenase(LDH).Reaction oxygen species(ROS)production was determined with DCFH-DA fluorescent probe.Malondialdelyde(MDA)was measured with spectroscopy and the anti-apoptosis protein Bcl-2 was measured with Western blot.Results PC12 cells were damaged by hydrogen peroxide.Compared with the model group,the survival rate of PC12 cells was improved significantly in morroniside groups.The content of LDH was significantly lower.Morroniside also prevented ROS injury induced by hydrogen peroxide and reduced MDA content.The anti-apoptosis protein Bcl-2 increased in morroniside groups.Conclusion Morroniside has protective effect on PC12 cells induced by hydrogen peroxide.
