RP-HPLC法测定肿节风分散片中异秦皮啶的含量

Determination of Isofraxidin Content in Sarcandrae Diskets by RP-HPLC

目的建立肿节风分散片中异秦皮啶的含量测定方法。方法采用反相高效液相色谱法对肿节风分散片中的主要成分异秦皮啶的含量进行测定,色谱柱为Diamonsil C18(250 mm×4.6 mm;5μm),流动相为乙腈-0.1%磷酸溶液20∶80(V/V);检测波长:344nm;流速:1ml·min^(-1);柱温:30℃。结果试验精密度RSD值为0.41%,r=0.9999,回收率为99.7%,重复性试验RSD值为1.19%,3批肿节风分散片中的异秦皮啶平均含量为1.073mg·g^(-1),稳定性良好。结论建立的方法简便易行、专属性强、准确度高,能有效检测肿节风分散片制剂中异秦皮啶的含量,可用于该药品在生产过程中的质量控制。

Objective To establish a method to determine the content of isofraxidin in sarcandrae diskets by RP-HPLC.Methods The chromatographic column was C18 column.The mobile phase consisted of acetonitrile and0.1% phosphoric acid solution at a ratio of 20∶80（V/V）.The UV detection wavelength was 344 nm,the flow rate 1 ml·min^-1,and column temperature was 30℃.Results The result showed that RSD of the experiment was0.41%（r=0.9999）,the recovery was 99.7%,while the RSD of the repetitive experiment was 1.19%.The isofraxidin content of three batches of samples was 1.073 mg·g^-1.Conclusion This method has the advantages of high specificity,accuracy and operability,which can be used to determine the isofraxidin content in sarcandrae disketsand control the quality of medicine in the process of production.
