补肾活血中药对薄型子宫内膜大鼠子宫内膜干细胞标志物的影响 被引量:8

Effects of Bushen Huoxue decoction on endometrial stem cell markers in rats with thin endometrium

摘要 目的探讨补肾活血中药对薄型子宫内膜大鼠子宫内膜干细胞标志物的影响。方法取处于动情期的雌性大鼠36只随机分为正常对照组、模型组、补佳乐组以及补肾活血中药高、中、低剂量组。除正常对照组外,其余各组大鼠建立肾虚血瘀薄型子宫内膜模型。正常对照组灌服蒸馏水,模型组建立模型后灌服蒸馏水,补佳乐组建立模型后灌服补佳乐0.3 mg/(kg·d),补肾活血中药高、中、低剂量组建立模型后分别灌服补肾活血中药8.64、4.32、2.16 mg/kg。经过3个动情周期后,处死大鼠,取材。免疫组织化学检测活化β-catenin蛋白的表达、RT-PRC检测Oct4基因表达以及Wertern blot法检测ABCG2蛋白的表达。结果 1)活化β-catenin蛋白的表达:模型组较对照组极显著降低(P<0.001),给药后各组β-catenin不同程度增加,低剂量组较模型组有显著性差异(P<0.05),中剂量组、高剂量组、补佳乐组与模型组有极显著性差异(P<0.001)。2)Oct4基因的表达:模型组较对照组极显著降低(P<0.001),给药后各组OCT4表达量不同程度增加,其中低剂量、中剂量与模型组无显著性差异,高剂量与模型组有极显著性差异(P<0.001),补佳乐组与模型组有显著性差异(P<0.05)。3)ABCG2蛋白的表达:模型组较对照组极显著降低(P<0.001),给药后各组ABCG2表达量不同程度增加,其中低剂量与模型组无显著性差异,中剂量组与模型组有显著性差异(P<0.05),高剂量、补佳乐组与模型组有极显著性差异(P<0.001)。结论补肾活血中药可以不同程度的增加β-catenin蛋白、OCT4基因及ABCG2蛋白的表达。 Objective To investigate the eff ect of Bushen Huoxue decoction on the markers of endometrial stem cells in rats with thin endometrium. Methods The estrous period 36 rats were randomly divided into normal control group, model group, Chinese medicine group and progynova High, medium and low dose groups. In addition to the normal control group, the other rats in each group established the thin endometrium model of kidney deficiency and blood stasis. The control group used distilled water, model group distilled water after model establishment, progynova orally after progynova 0.3 mg/(kg·d), the traditional Chinese medicine for tonifying kidney and promoting blood circulation in the high, middle andlow dose groups were respectively administrated with Bushen Huoxue traditional Chinese medicine 8.64, 4.32, 2.16 mg/kg. After 3 estrous cycles, the rats were sacrificed. Activation of beta was detected by immunohistochemistry-catenin protein expression, RT-PRC detection, Oct4 gene expression, and wertern blot method to detect ABCG2 protein. Results 1. Activation of the expression of β-catenin protein: the model group was significantly lower than the control group(P〈0.001). The levels of beta-catenin in each group increased, and the low dose group had significant difference compared with the model group(P〈0.05). The medium,high and progynova groups were significantly higher than the model group(P〈0.001). 2. Expression of Oct4 gene: compared with model group,the control group was significantly lower(P〈0.001). After the administration, the expression of OCT4 in each group increased to a certain extent. There was no significant diff erence with the model group, there was significant diff erence between the high dose group and model group(P〈0.001), progynova group and model. There was significant diff erence between the two groups(P〈0.05). 3. Expression of ABCG2 protein: the model group was significantly lower than that of the control group(P〈0.001). After administration, the expression of ABCG2 in each group increased to some extent, and there was no significant diff erence between the low dose and the model group.Type group had significant diff erence(P〈0.05), the high dose, there was significant diff erence between the two groups and the model group(P〈0.001). Conclusion Bushen Huoxue decoction can somehow increase the expression of beta-catenin protein, OCT4 gene and ABCG2 protein.
