水泥粉尘对小鼠肺脏的影响

摘要 目的探讨水泥粉尘对小鼠肺脏组织的影响。方法将24只雄性清洁级ICR小鼠参照随机原则分为4组:对照组(不染毒)、实验1组(染毒7天)、实验2组(染毒14天)、实验3组(染毒28天),每组6只。采用静式染毒法对小鼠进行染毒。染毒结束并禁食24 h后测定脏体比、血清过氧化物歧化酶(SOD)、肺组织羟脯氨酸(Hyp)含量,并观察肺组织病理学改变。结果 (1)脏体比:与对照组进行比较,实验2组脏器系数明显下降(P<0.05),与实验2组进行对比,实验3组脏器系数增高(P<0.05);(2)血清SOD活力:与对照组相比较,实验1组SOD活力明显提高(P<0.05),与实验1组相比较,实验2组SOD活力明显下降(P<0.05),实验3组与实验2组比较,差异无统计学意义;(3)肺组织Hyp含量:与对照组比较,实验1组羟脯氨酸含量明显降低(P<0.05),实验2组羟脯氨酸含量明显降低(P<0.05),与对照组、实验1组、实验2组比较,实验3组羟脯氨酸含量明显升高(P<0.05);(4)病理学检查结果:实验3组小鼠肺组织切片中,入股坏死脱落的支气管壁粘膜上皮细胞粘附黑色粉尘颗粒,肺泡内的巨噬细胞数量少,个别细胞疑似吞噬有粉尘颗粒。结论水泥粉尘暴露能够引起小鼠的肺组织氧化损伤,并有时间关联。接触一定时间后,可导致小鼠肺组织胶原纤维升高,提示纤维化进程的开始。 [Objective] To explore the effect of cement dust on lung of mice. [Methods] Totally 24 male ICR mice were randomly divided into 4 groups(n=6): control group(no dust), the experimental group 1 (7 days), the experimental group 2 (14 days) ,the experimental group 3 (28 days). Then the mice in experimental groups were exposed to cement dust under static inhalation. After exposure and fasting for 24 h, the organ-body ratio, concentrations of SOD and Hyp were examined, pulmonary pathological changes were observed. [Results] ①The organ-body ratio: compared with the control group, the levels of organ coefficient and Hyp in the experimental group 2 were significantly decreased (P〈0.05). There was statistical significance of the organ coefficient between the experimental group 2 and the experimental group 3 (P〈0.05). ②The vitality of serum SOD: compared with the control group, the level of SOD in the experimental group 1 was significantly creased (P〈O.05). The SOD in experimental group 2 was significantly decreased, compared with group 1. There was no significantly difference between group 3 and group 2. ③The Hyp content of lung tissue: compared with control group, the Hyp level in experimental group 1 was significantly decreased (P〈O.05). The level of Hyp in the experimental 3 was statistically creased than other 3 groups (P〈0.05). ④Pathological results: black dust particles adhered to epithelial cells in the lining of the bronchial wall and there were a few maerophages in the alveoli. Individual cells might swallowed dust particles. [Conclusion] The exposure of cement dust can cause oxidative damage in mice, with time correlation. After exposure for a certain period, collagen fiber will elevate in lung tissues of mice which indicates the beginning of the fibrosis orocess.
