解郁开心丹对抑郁模型大鼠海马及缰核中BDNF表达的影响 被引量:4

摘要 目的观察解郁开心丹对抑郁模型大鼠海马及缰核中BDNF表达的影响。方法将60只SD雄性大鼠随机分为正常组、模型组、西药组、中药高、中、低剂量组,共6组,每组10只,使用慢性应激结合孤养的方法造模。给药,西药组盐酸氟西汀10 mg/d;中药高、中、低组分别给予2.0、1.0、0.5 g/d的解郁开心丹中药汤剂,连续28 d。用免疫组化法检测大鼠海马及缰核中BDNF表达。结果与模型组比较,中药高、中组大鼠海马及缰核中BDNF的表达升高(P<0.05);中药高、中剂量组与西药组比较,差异无统计学意义(P>0.05)。结论解郁开心丹与西药氟西汀对抑郁模型大鼠海马及缰核中BDNF的表达具有同样的调节作用,其抗抑郁作用机制可能与调节大鼠海马及缰核中BDNF的表达有关。 Objective To observe the effect of Jieyu Kaixin Dan on the expression of BDNF in the hippocampus and rein nuclear in rats with depression model. Methods The 60 SD male rats were randomly divided into normal group, model group, western medicine group, Chinese medicine high, medium and low dose group, with 10 in each group. Chronic stress combined with solitary care was used to model. The western medicine group was given fluoxetine hydrochloride 10 mg/d, and the high, middle and low groups were given the decoction of Jieyu Kaixin Danof 1.0-0.5 g/d for 28 days. Immunohistochemistry was used to detect the expression of BDNF in the hippocampus and rein nuclear in rats. Results Compared with the model group, the expression of BDNF in the hippocampus and rein nuclear of the rats in the high and middle group was increased(P〈0.05), Chinese medicine high, middle dose group and western medicine group have no statistical significance(P〉0.05). Conclusion It was found that the expression of BDNF in the hippocampus and rein nuclear in rats with depression model was the same as that of the fluoxetine. Therefore, the mechanism of the antidepressant effect may be related to the regulation of BDNF expression in the hippocampus and habenula in rats.
