疏肝调神针法对偏头痛大鼠行为学及血中相关神经肽、炎性物质表达的影响 被引量:25

Effect of Manual Acupuncture Preconditioning on Behavior and Contents of Serum CGRP,SP,IL-1β and TNF-α Levels in Migraine Rats

摘要 目的:观察疏肝调神针法对偏头痛大鼠的行为学及血清降钙素基因相关肽(CGRP)、P物质(SP)、白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)表达的影响,探讨疏肝调神针法治疗偏头痛可能的作用机制。方法:Wistar雄性大鼠随机分为对照组、模型组、疏肝调神组、普通针刺组,每组8只。疏肝调神组针刺"百会"及双侧"风池""内关""太冲",普通针刺组针刺"百会"及双侧"风池",30min/次,1次/d,连续8d。第8天针刺后采用大鼠颈后皮下注射硝酸甘油制备偏头痛模型。观察造模前后3次行为学评分变化,酶联免疫法检测各组血清中CGRP、SP、IL-1β、TNF-α的浓度。结果:造模后,各组行为学评分均较对照组明显升高(P<0.05);造模后2个评分时间段(60~90min及120~150min),与模型组比较,疏肝调神组、普通针刺组行为学评分明显降低(P<0.05);120~150min时,疏肝调神组与普通针刺组评分较60~90min时明显降低(P<0.05)。与对照组比较,模型组大鼠血清中CGRP、SP、IL-1β、TNF-α的浓度明显升高(P<0.05);与模型组比较,疏肝调神组、普通针刺组大鼠血清中CGRP、SP、IL-1β、TNF-α的浓度明显降低(P<0.05);与普通针刺组比较,疏肝调神组大鼠血清中CGRP、SP、IL-1β、TNF-α的浓度显著降低(P<0.05)。结论:针刺可有效防治偏头痛的发生,疏肝调神针法在抑制偏头痛大鼠血清CGRP、SP及IL-1β、TNF-α等因子的痛觉传导、减轻神经炎性反应中更具优势。 Objective To observe the effect of liver-soothing and mental-activity-regulating (LSMAR) need behavior reactions and contents of serum calcitonin gene-related peptide (OGRP), substance P (SP), interleukin-1β (IL-1 tumornecrosis factor-α (TNF-a) in migraine rats, so as to explore its mechanism underlying relief of migraine. Methods mg on 15) and Thirty- two male Wistar rats were randomly divided into control, model, LSMAR needling and conventional needling groups (n = 8 rats in each). The migraine model was established by hypodermic injection of glyceryl trinitrate (5 mg/kg) at the napex. Acupuncture preconditioning was conducted once daily for 8 days before modeling. Acupuncture needles were inserted into "Baihui" (GV 20), and bilateral "Fengchi" (GB 20), "Neiguan" (PC 6) and "Taichong" (LR 3), manipulated for a while and retained for 30 min. The rats' behavioral changes (times of head scratching, tail-biting, cage-crawling, and to- and fro-movement in 30 min) werescored (one symptom = one point) before modeling, after modeling, and after intervention. The concentrations of OGRP, SP, IL-1β, TNF-α in serum were assayed by enzyme-linked immunosorbent assay (ELISA). Results After modeling, the behavioral scores were significantly increased in the migraine model rats relevant to the control group ( P〈0.05). Whereas after the treat- ment, the behavior scores were considerably decreased during 60 - 90 min and 120 - 150 min of the measuring period in both LSMAR and conventional needling groups compared with the model group ( P〈0.05). The concentrations of CGRP, SP, IL-1β and TNF-α were significantly higher in the model group than in the control group ( P〈0.05), and notably lower in both LSMAR and con- ventional needling groups than in the model group ( P〈0.05). The therapeutic effect of LSMAR was markedly superior to that of conventional needling in down-regulating the concentrations of OGRP, SP, IL-1β and TNF-α (P〈0.05). Conclusion Acupunc- ture preconditioning can effectively relieve pain in migraine rats possibly by decreasing the concentrations of OGRP, SP, IL-1β and TNF-αin the peripheral blood, and the therapeutic effect of LSMAR is evidently superior to that of conventional needling.
