敲除FGF5基因对陕北白绒山羊血液生理生化指标的影响 被引量:5

Determination and Analysis of FGF5 Gene Knockout on the Physiological and Bio-Chemical Parameters of the Blood of Shanbei Cashmere Goats

摘要 为探究敲除FGF5基因对陕北白绒山羊血液生理生化指标的影响,本实验以敲除FGF5基因的陕北白绒山羊(公、母羊各8只,阳性)、同龄普通陕北白绒山羊(公、母羊各5只,阴性)为研究对象,分别在10月龄、1岁与1.5岁时测定8项血常规指标和14项血液生化指标。结果表明:血常规指标中,10月龄、1岁时,阳性公羊的红细胞压积(HCT)、平均红细胞血红蛋白含量(MCH)显著低于阴性公羊(P<0.05),阳性母羊的红细胞分布宽度变异系数(RDW)、血红蛋白(HGB)、HCT极显著高于阴性母羊(P<0.01);1岁时,阳性公羊和母羊的红细胞数目(RBC)均显著高于阴性公羊和母羊(P<0.05);1.5岁时,阳性母羊的白细胞数目(WBC)显著高于阴性母羊(P<0.05)。血液生化指标中,10月龄时,阳性母羊的球蛋白极显著低于阴性母羊(P<0.01);1岁、1.5岁时,阳性母羊的白蛋白极显著高于阴性母羊(P<0.01);1.5岁时,阳性母羊的尿素极显著低于阴性母羊(P<0.01);其余指标均差异不显著(P>0.05)。因所有指标均在参考范围内,说明敲除FGF5基因的陕北白绒山羊的蛋白质代谢、糖代谢、脂类代谢、肝胆功能、钙磷代谢均无显著影响。表明采用CRISPR/Cas9技术敲除FGF5基因并未对陕北白绒山羊的健康产生影响,但部分存在显著差异的血液生理生化指标有待进一步研究。 The experiment was conducted to study the effect of FGF5(Fibroblast Growth Factor-5) gene knockout on the physiological and bio-chemical parameters of the blood of Shanbei cashmere goats. The experimental animals were 8 FGF5-knockout female Shanbei cashmere goats(knockout female group), 8 FGF5-knockout male Shanbei White cashmere goats(knockout male group), 5 female Shanbei White cashmere goats(control female group) and 5 male Shanbei cashmere goats(control male group).The levels of WBC, RBC, HGB, HCT, MCV, MCH, MCHC and RDW in the blood of Shanbei White cashmere goats at the age of 10, 12 or 18 month old were determined, meanwhile the activities of AMY, ALT, ALP and CK, the levels of TP, ALB, GLO, Ca, GLU, Urea, P, TC, TBIL and CREA in the blood were measured too.Results: The results showed that the blood routine parameters were not significant(P0.05) except for RDW(P0.05) of the knockout and the control groups, while all the parameters were within the normal range. The blood routine parameters differed within the normal value among Shanbei cashmere goats of different ages and genders. The blood biochemical indexes were different of Shanbei cashmere goats of different ages and genders but they were within the normal range. The protein metabolism, glucose and lipid metabolism, liver and gallbladder function, metabolism of calcium and phosphorus of the knockout FGF5 Shanbei White cashmere goats had no significant difference.Conclusion: The blood biochemical indexes were different of Shanbei white cashmere goats of different ages and genders but they were within the normal range. There were no significant difference between the knockout group and the control group. The results demonstrated that CRISPR/Cas9-edited FGF5 gene had no significant difference on the blood biochemical indexes of Shanbei cashmere goats.
