电针背俞穴对慢性疲劳大鼠下丘脑CRHmRNA表达及血清ACTH、CORT的影响

Effect of "Beishu Points" Electroacupuncture Stimulation on CRHmRNA, ACTH and CORT in Rats with Chronic Fatigue Sydome

目的:通过研究电针背俞穴对慢性疲劳综合征(CFS)大鼠下丘脑CRHmRNA、血清促肾上腺皮质激素(ACTH)、血清皮质醇(CORT)的影响,探讨针刺治疗CFS的作用机制。方法:雄性SD大鼠45只随机分为空白组、模型组、电针组,每组15只。采用睡眠剥夺+束缚+负重游泳的复合应激法制备模型。电针组选穴:心俞、肺俞、脾俞、肝俞、肾俞,于针尾加电,强度为1 m A,频率为2 Hz,4个疗程后(1次/d,5 d为1个疗程,疗程间休息2 d)测量其CRHmRNA表达情况及血清ACTH、CORT含量,分别采用qPCR及酶联免疫法检测。结果:干预后,模型组大鼠体质量低于电针组及空白组(P<0.05);电针组大鼠血清ACTH、CORT含量及下丘脑CRHmRNA表达均低于模型组(P<0.05)。结论:电针可调节CFS大鼠下丘脑CRHmRNA表达及血清ACTH、CORT水平,可能是电针治疗CFS的作用机制之一。

Objective： To observe the effect of ＂Beishu points＂ electroacupuncture stimulation on CRHmRNA, Serum Adrenocorticotropic hormone （ACTH） and Cortisol （CORTP in rats with chronic fatigue sydome （CFS）. Methods： A total of 45 male SD rats were equally randomized into control group, model group and electric acupuneture group, 15 rats in each. CFS modal was established by bounding and sleep deprivation with load swimming. Rats in the electroacupuncture group were treated by electroacupuncture stimulation of ＂Xinshu （BL15） Feishu （BL13） Pishu （BL20） Ganshu （BL18） Shenshu （BL23）＂ with the intensity of 1 mA and the frequeney of 2 Hz. The CRHmRNA, ACTH and CORT were detected by qPCR and Elisa after 4 courses （1 timesMay, 5 days for a course of treatment, rest for 2 days between two courses of treatment）. Results： The model group showed lower weigh than the normal and electric acupuncture groups （P〈0.05）; The electric acupuncture group showed lower ACTH, CORT and CRHmRNA expression than the model group （P〈0.05）. Conclusion： Electroacupuncture can regulate the level of CRHmRNA, ACTH and CORT in CFS rats, which may be one of the mechanisms of EA treatment of CFS.
