摘要
采用丙酮/柠檬酸三钠两相盐析体系提取沙棘果渣中多酚、类黄酮和原花青素。考察溶剂与盐用量、目数、时间、料液比和温度对有效成分得率的影响,并用响应面法对提取工艺进行进一步优化。结果表明:目数为160目,料液比1︰45(g/m L),静置温度35℃,静置时间88 min,体系组成为27%丙酮/14%Na3C6H5O7时,多酚、类黄酮及原花青素富集于上相溶剂相,其得率和回收率有最大值,分别为57.56,5.75,43.19 mg/g和96.63%,91.10%,98.87%。与其他提取方法相比,双水相提取法溶剂用量少,提取时间短,所的提取物具有更高的得率及抗氧化性能。
The acetone/Na_3C_6H_5O_7 salting-out two-phase system(SOTPS) was employed to extract polyphenols, flavonoids and proanthocyanins from sea buckthorn marc. The influences of solvent and salt dosage, particle size, time, material-liquid ratio and temperature on the extraction rate of polyphenols, flavonoids and proanthocyanins were investigated. Then the extraction process was optimized by response surface method. When SOTPS consisted of 27% acetone/14% Na_3C_6H_5O_7, the extraction rate of polyphenols, flavonoids and proanthocyanins and the recovery rate were 57.56, 5.75, 43.19 mg/g and 96.63%, 91.10%, 98.87%, respectively, under the optimum condition of sample number 160 mesh, solid-liquid ratio 1︰45(g/m L), the static temperature of 35 ℃, static time 88 min. Compared with other extraction methods, aqueous two-phase extraction has advantage of solvent, shorter extraction time, better yield and antioxidant properties.
