摘要
目的建立大鼠血浆中右旋布洛芬浓度的测定方法。方法血浆经乙腈沉淀蛋白后,取上清液检测,处理;色谱分析采用Zorbax Eclipse XDB-C8(150 mm×4.6 mm,5 m);流动相为甲醇:0.01mmol/L磷酸二氢钾(75∶25),用磷酸调至p H=3.6,流速1.0m L/min;柱温25℃,检测波长220nm。结果右旋布洛芬在15.6~500 g/m L范围内呈良好的线性关系,回归方程Y=1.9024X-3.358,R=0.9987。结论建立的方法准确精密、稳定可靠,可用于大鼠血浆中右旋布洛芬浓度的测定。
Objective To establish a method for determination of dexibuprofen concentration of rat plasma. Methods After the plasma was precipitated with acetonitrile,the supernatant was detected and treated. The chromatographic analysis used Zorbax Eclipse XDB-C8(150 mm× 4.6 mm,5 m),and the mobile phase was methanol,0.01 mmol/L phosphate dihydrogen potassium(75∶25),phosphoric acid to p H=3.6,flow rate was 1 m L/min,column temperature was 25℃,and wavelength was220 nm. Results It showed a good linear relationship of dextroibuprofen in the range of 15.6~500 g/m L,and the regression equation was Y=1.9024 X-3.358,R=0.9987.Conclusion The established method is accurate,stable and reliable,and can be used for the determination of dexibuprofen in rat plasma.
