补肾消石退黄颗粒中绿原酸的含量测定

Content Determination of Chlorogenic Acid in Bushen Xiaoshi Tuihuang Granutes

目的建立补肾消石退黄颗粒中绿原酸含量的HPLC测定方法。方法色谱柱:Kromasil C18(5 m,4.6 mm×250 mm);流动相:乙腈-0.1%磷酸溶液(10∶90);流速:1.0m L/min;柱温:30℃;检测波长:327 nm。结果绿原酸在0.07872~0.9446 g范围内有较好的线性关系(r=0.9999),平均加样回收率为97.36%(RSD=2.96%)。结论该方法重复性好、灵敏度高,操作简便易行,可用于测定补肾消石退黄颗粒中绿原酸的含量。

Objective To establish a method for content determination of chlorogenic acid in Bushen Xiaoshi Tuihuang granutes by HPLC. Methods On the column of Kromasil C18（5 m,4.6 mm×250 mm）with the mobile phase of acetonitrile-0.1%phosphoric acid solution（10∶90）at the flow rate of 1.0 m L/min,the detection wavelength of 327 nm,the temperature of 30℃.Results The peak area of chlorogenic acid showed good linear relationship（r=0.9999）within the concentrations of 0.07872-0.9446 g. The average recovery rate was 97.36%（RSD=2.96%）. Conclusion The established HPLC method is with good repeatability,high sensitivity,simple operation,and can be applied in the content determination of chlorogenic acid in Bushen Xiaoshi Tuihuang granutes.
