连续色谱法分离甘蔗叶提取物制备阿拉伯糖和木糖的研究 被引量:3

摘要 [目的]建立一种高效提取甘蔗叶木糖母液中L-阿拉伯糖和D-木糖的方法。[方法]以PCR-642 Ca^(2+)树脂为固定相,纯水为流动相,使用连续色谱分离移动床对关键组分L-阿拉伯糖和D-木糖进行了分离,探讨了Ⅰ区、Ⅱ区、Ⅲ区、Ⅳ区流量、进料流速、进料浓度等操作条件对连续移动床色谱分离效果的影响。[结果]结果表明最佳工艺参数为:进料浓度44.12%,进料流速10.0mL/min,Ⅰ区流速为73.0mL/min,Ⅱ区流速为60.0mL/min,Ⅲ区流速为70.0mL/min,Ⅳ区流速为57.0mL/min,L-阿拉伯糖出口流速13.0mL/min,木糖出口流速15.0mL/min。在此条件下L-阿拉伯糖出口浓度为9.37%,纯度达到90.41%,收率达到93.69%;D-木糖出口浓度为20.79%,纯度达到92.07%,收率达到92.12%。[结论]试验为寻找一条利用甘蔗叶木糖母液等富含D-木糖、L-阿拉伯糖的料液,来生产L-阿拉伯糖和D-木糖的高效、环保、成本低、易于实现的生产工艺提供了参考。 [Objective] The aim was to establish high efficiency method of extracting L-arabinose and D-xylose from the xylose crystalline mother liquid of sugarcane leaves. [Method] With resin of PCR-642 Ca^(2+)as the padding and deionized water as the mobile phase, continuous chromatographic separator bed was used to separate the key components of L-arabinose and D-xylose. The influences of operating conditions(flow rate in sectionⅠ,Ⅱ, Ⅲ, Ⅳ, feed flow rate,and feed concentration) on separation effects of the continuous chromatographic separator bed were studied. [Result] The optimized parameters were: feed concentration 44.12%, feed rate 10.0 mL/min, flow rate in sectionⅠ 73.0 mL/min, flow rate in sectionⅡ 60.0 mL/min, flow rate in section Ⅲ 70.0 mL/min, flow rate in section Ⅳ 57.0 mL/min, L-arabinose rate 13.0 mL/min and D-xylose rate 15.0 mL/min. Under these conditions, the concentration of purified L-arabinose was 9.37% with the purity of 90.41% and the recovery was 93.69%, and the concentration of purified D-xylose was 20.79% with the purity of 92.07% and the recovery was 92.12%. [Conclusion] This study provides a reference in search of the xylose crystalline mother liquid from extraction of sugarcane leaves enriched in D-xylose liquid and L-arabinose liquid, to produce L-arabinose and D-xylose with efficiency, environmental protection, low cost and easy to realize production process.
