心房钠尿肽下调呼吸道合胞病毒毛细支气管炎小鼠肺组织T-bet水平 被引量:1

摘要 目的研究T-bet在呼吸道合胞病毒毛细支气管炎小鼠肺组织中的表达及心房钠尿肽刺激下T-bet的变化。方法建立毛细支气管炎小鼠模型,观察心房钠尿肽的变化;同时利用肺组织病理学、ELISA法和实时定量PCR法检测心房钠尿肽干预后气道炎症浸润、T-bet mRNA及蛋白的表达和IFN-γ等的变化。结果与正常对照组相比,呼吸道合胞病毒组小鼠肺组织中心房钠尿肽水平明显升高;而经鼻吸入心房钠尿肽可使气道炎症细胞浸润加剧,T-bet及IFN-γ表达水平均降低;而利用心房钠尿肽拮剂预处理后,心房钠尿肽的上述效应可被明显逆转。结论心房钠尿肽在毛细支气管炎肺组织中存在一定量的表达,同时可下调T-bet水平。 This study was designed to study the expression of T-bet in the lung tissue and its changes under the stimulation of atrial natriuretic peptide (ANP) in mice with respiratory syncytial virus (RSV) bronehiolitis. mouse model of bronchiolitis was established to observe the changes of airway inflammatory infiltration, the expression of T-bet mRNA and IFN-γ in lung tissue by using the method of histopathology, ELISA and real-time quantitative PCR. Compared with the normal group, the level of ANP in the lung tissue of the RSV group was significantly increased. Nasal inhalation of ANP can severely increase the infiltration of airway inflammatory cells and decrease the expression of T-bet and IFN-γ. While, pretreatment of ANP agonist can significantly reverse the effects mentioned above. Taken together, ANP can be expressed in lung tissue of bronchiolitis, besides, it can down regulate the level of T-bet.
