摘要
为了提高红树莓籽油提取率,以红树莓籽为原料,采用胃蛋白酶、酸性纤维素酶、果胶酶、a-淀粉酶、胰蛋白酶、碱性蛋白酶提取红树莓籽油工艺优化,在单因素实验的基础上,根据酶的理化特性,分别对酸性复合酶与碱性复合酶提取油脂进行研究,采用Box-Behnken法优化两组复合酶提取工艺参数。结果表明:酸性复合酶最佳提取工艺参数:胃蛋白酶添加量0.76%,果胶酶添加量1.51%,酸性纤维素酶添加量1.07%,提油率(86.11±0.09)%;碱性复合酶最佳提取工艺参数:胰蛋白酶添加量1.52%,碱性蛋白酶添加量1.99%,α-淀粉酶添加量2.52%,提油率(88.75±0.08)%。结论:碱性复合酶提油率高于酸性复合酶。
To improve the extraction rate of red raspberry seed oil,red raspberry seeds were used as raw material to optimize the extraction processing by using pepsin,acid cellulase,pectinase,a-amylase,trypsin and alkaline protease. Based on the single factor test,the effect of acid compound enzyme and alkaline compound enzyme on extraction of red raspberry seed oil were studied according to the physical and chemical properties of the enzyme. The Box-Behnken method was used to optimize the extraction parameters of the two groups. The results showed that the best extraction parameters of acid compound enzyme were as follows:pepsin addition amount 0. 76%,pectinase addition amount 1. 51%,acid cellulase addition amount 1. 07%,oil extraction rate( 86. 11 ± 0. 09) %. The best extraction parameters of alkaline compound enzyme were as follows:trypsin addition amount 1. 52%,alkaline protease addition1. 99%,α-amylase addition 2. 52%,oil extraction rate(88. 75 ± 0. 08)%. In conclusion,the extraction rate of alkaline compound enzyme is higher than that of acid complex enzyme.
