剥脱性点阵激光联合射频对大鼠皮肤的影响 被引量:3

摘要 目的:探讨剥脱性点阵激光联合射频对大鼠皮肤的影响,对二者联合治疗机制进行初步研究。方法:40只SD大鼠背部脱毛后,将其背部划分为4个区域:A组(空白对照组)、B组(CO2点阵激光组)、C组(多极射频组)、D组(CO2点阵激光联合多极射频组)。分别于每组干预完全结束后第2W、4W、8W、12W进行取材,予HE染色、Masson染色、Ⅲ型胶原纤维免疫组化、VEGF免疫组化。结果:HE染色显示:干预后第8W、12W,各干预组均可见明显新生胶原、排列致密,D组增厚最明显(P<0.05);Masson染色显示干预后第8W、12W,D组胶原纤维更致密(P<0.05);免疫组化Ⅲ型胶原纤维显示干预后第2W,各干预组Ⅲ型胶原纤维合成增多,干预后第4W,C组、D组持续增多,D组更显著(P<0.05);免疫组化VEGF显示干预后第2W、4W、8W,与A组比较,其余各干预组平均光密度值明显增大(P<0.05),干预后第12W,C组、D组表达仍强于A组(P<0.05)。结论:剥脱性点阵激光联合多极射频治疗对促进真皮胶原纤维合成的作用更显著;多极射频、剥脱性点阵激光联合多极射频较单独应用剥脱性点阵激光使真皮血管内皮细胞VEGF表达上调持续时间长。 Objective: To investigate the effects of ablative fractional laser combined with radiofrequency on the rat skin and its underlying mechanisms. Methods: In a total of 40 SD rats, the depilated back of each rat was divided into four ar- eas: A(blank control), B(CO2 fractional laser), C(muhipolar radiofrequency), and D(CO2 fractional laser+muhipolar radiofre- quency). Following treatments, skin samples were collected at week 2, week 4, week 8 and week 12 for H&E staining, Mas- son staining, and staining of type Ⅲ collagen and VEGF. Results: H&E staining showed increased new collagen in all treated areas, particularly in area D, at both weeks 8 and 12(area D vs. others, P〈0.05). Masson staining al- so demonstrated a dramatic increase in collagen densities in area D at both weeks 8 and 12 after intervention (area D vs. others, P〈0.05). Immunohistochemical staining showed that all treatments increased typeⅢcollagen at week 2, with a fur- ther increase in areas C and D at week 4(area D vs. others, P〈0,05). In comparison to area A, average optical densities of VEGF staining increased significantly in all treated areas 2 to 8 weeks after treatments(area A vs. others, P〈0.05). Average optical densities of VEGF staining were still higher in areas C and D than in area A 12 weeks after treatments (area A vs. C & D, P〈0.05). Conclusions: Ablative fractional laser combined with multipolar radiofrequency ablation is more effective in promoting the synthesis of dermal collagen fibers. The combination of multipolar radiofrequency ablation with ablative fraction- al laser induces a sustained increase in VEGF expression in dermal vascular endothelial cells.
