摘要
目的观察三伏灸治疗对支气管哮喘新西兰兔大椎穴和肺俞穴透析液腺苷酸含量及肺组织P2X7受体m RNA表达的影响。方法选取新西兰雄性兔子18只,随机分为3组。进行支气管哮喘气道重塑模型造模后,于"三伏天"以隔姜灸加中药贴敷法干预,疗程结束后,采用微透析的方法提取大椎穴、肺俞穴的透析液,再用超高效液相色谱法测定透析液中腺苷酸的含量。取新西兰兔肺组织,采用荧光定量PCR法观察各组肺组织P2X7受体m RNA表达的变化。结果与空白组相比,模型组大椎穴和肺俞穴的ATP浓度均明显升高(P<0.05),与模型组相比,治疗组的大椎穴和肺俞穴的ATP浓度均明显下降(P<0.05);模型组新西兰兔肺组织P2X7受体m RNA的表达较空白组明显升高(P<0.05),治疗组P2X7受体m RNA的表达较模型组明显下降(P<0.05)。结论 ATP可能通过P2X7受体参与了支气管哮喘的发生发展过程,三伏灸可通过下调局部穴位ATP的浓度,进而调整肺组织P2X7受体的表达量,达到治疗效果。
Objective To observe the effect of ATP and P2 X7 expression about New Zealand white rabbits which treat by dog days moxibustion. Methods 18 male New Zealand white rabbits were randomly divided into blank group, model group and experimental group. Airway remodeling of asthma model, experimental group were given medicine applicator Ginger joint intervention groups Ginger and sticking points are selecting Dazhui( GV14) and bilateral Feishu( B13). At the end of the experiment, extrac the dialysate of Dazhui( GV14) and Feishu( B13) by microdialysis method, and the ATP content was detected. The rabbits lung tissue was took, and the P2 X7 gene expression was detected. Results Comparing with the blank group, in the model group the ATP content of Dazhui( GV14) and Feishu( B13) were significantly increased( P 0.05). Comparing with the model group, in the experimental group the ATP content of Dazhui( GV14) and Feishu( B13) were significantly decreased( P 0.05). Comparing with the blank group, the gene expression of P2 X7 were significantly increased in the model group( P0.05). Comparing with the model group, the gene expression of P2 X7 were significantly decreased in the experimental group(P0.05). Conclusion ATP may be involved in the development of bronchial asthma by P2 X7. Dog days moxibustion may be decrease the ATP content and then decrease the gene expression of P2 X7 to achieve the effect of treatment.
