摘要
目的探讨微小核糖核酸(microRNA,miR)-486-5p在骨髓增生异常综合征(MDS)中的表达水平,为研究miR-486-5p在MDS疾病发生发展中的作用提供参考。方法利用分光光度计及变性凝胶电泳对9例MDS患者(病例组)和6例正常造血干细胞移植术供者(对照组)骨髓标本进行质量检测,然后运用基因表达谱芯片结合分层聚类从病例组和对照组骨髓中筛选出有显著差异表达的miR-486-5p,进行两组miR-486表达水平的比较。结果相比健康对照组,miR-486-5p在MDS患者骨髓中表达量(中位直径标准化后表达量)显著上调(4.10±1.88 vs0.91±0.53,r=4.51,t=3.997,P=0.002)。结论基因芯片结果显示miR-486-5p在MDS患者骨髓中表达显著上调,其可能通过靶向负调控下游的靶基因或调控其信号转导通路而参与MDS疾病的发生发展,降低miR-486-5p的表达水平有可能成为MDS的治疗手段。
Objective To investigate the expression level of microRNA (miR)-486-Sp in myelodysplastic syndrome (MDS) in order to provide theoretical basis for the effect of miR-486-Sp in the occurrence and development of MDS. Methods The bone marrow specimens of 9 MDS patients (case group ) and 6 donors of hematopoietie stem cell transplantation (control group) were selected. Spectrophotometer and denatured gel electrophoresis were used to carry out quality inspection,and then gene expression profiling mieroarray and hierarchical cluster analysis were used to screen out MiR-486-Sp with difference expression for the comparison of their expression levels between two groups. Results Genechip showed that miR-486-Sp expression (median expression level post normalization) in bone marrow of MDS patients was up-regulated significantly compared with control group (4. 10 ± 1.88 vs 0. 91 ± 0. 53, r = 4.41, t = -3. 997, P =0. 002). Conclusions miR-486-Sp expression in bone marrow of MDS patients is significantly up-regulated maybe through negative regulation of downstream target gene or its signal transduction pathway so as to participate in the occurrence and development of MDS. Reducing the expression level of miR-486-Sp may be a therapeutic method for MDS.
