姜黄素对大鼠环孢素A肾间质纤维化的保护作用

Protective effect of Curcumin on the renal interstitial fibrosis of rat by Cyclosporin A

目的探讨姜黄素(Cur)对环孢素A(Cs A)而致的大鼠肾间质纤维化的保护作用及其可能的分子机制。方法将18只SD大鼠随机分为对照组(Ctrl组,n=6)、Cs A组(n=6)和Cs A+Cur组(n=6),分别给予橄榄油、Cs A、Cs A联合Cur处理。通过测量大鼠的体重、取血测尿素氮和肌酐、肾组织病理学检查等指标观察Cur对动物肾间质纤维化的改善作用。通过检测大鼠肾组织超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量反映其ROS水平。采用分子生物学方法检测肾组织转化生长因子-β(TGF-β)的表达情况。结果 Cs A组动物相对于Ctrl组,体重下降(P<0.05),血尿素氮和肌酐水平显著上升(P<0.05),肾组织TGF-β表达上调(P<0.05),ROS水平上升,病理学表现为轻度间质纤维化,但同时给予Cur可以在一定程度上缓解上述表现。结论 Cur对Cs A而致的大鼠肾间质纤维化具有一定的保护作用,其分子机制可能是降低体内ROS水平和下调TGF-β表达。

Objective To investigate the protective effects of Curcumin（Cur） on the renal interstitial fibrosis induced by Cs A and reveal the underlying molecular mechanisms.Methods 18 SD rats were randomly divided into control group（Ctrl group,n=6）,Cs A group（n=6） and group Cs A＋Cur group（n=6） and the above-mentioned groups was treated with olive oil,Cs A and Cs A combined with Cur respectively.The effect of Cur on renal interstitial fibrosis in rats were observed by measuring body weight,blood urea nitrogen and creatinine,and histopathological examination.The level of ROS was detected by detecting SOD activity and MDA content in rat kidney tissue.The expression of transforming growth factor beta（TGF-β） in renal tissue was detected by molecular biology.Results Compared with ctrl group,weights of rats in Cs A group decreased obviously（P〈0.05）,and the level of blood urea nitrogen and creatinine increased significantly（P〈0.05）,and the expression of TGF-β increased significantly（P〈0.05）.The level of ROS increased, and the pathological findings were mild interstitial fibrosis.However,giving Cur at the same time could alleviate the above symptoms to some extent.Conclusion Cur has a some protective effect on the renal interstitial fibrosis which induced by Cs A,and its molecular mechanism may be down-regulating the expression of TGF-β and the level of ROS.
