NB-UVB治疗前后银屑病皮损中HSPs家族表达的变化及其与炎症反应的相关性 被引量:1

The changes of HSPs family expression in psoriasis lesions and their correlation with inflammatory response before and after NB-UVB treatment

摘要 目的:研究窄谱中波紫外线(NB-UVB)治疗前后银屑病皮损中HSPs家族表达的变化及其与炎症反应的相关性。方法:选择我院皮肤科首次诊断为寻常型银屑病并接受治疗的病例,随机分为接受NB-UVB联合常规治疗的NB-UVB组、接受常规治疗的对照组。治疗前及治疗后20天时,取银屑病皮损组织适量并测定HSPs家族分子、淋巴细胞转录因子的表达量以及炎症细胞因子的含量。结果:与治疗前比较,两组治疗后银屑病皮损组织中HSP27、HSP60、HSP90α、HSP90β、T-bet、Foxp3的mRNA表达量以及MCP-1、IFN-γ、IL-17的含量均显著降低,GATA-3、RORγt的mRNA表达量以及IL-10、TGF-β1的含量均显著升高且NB-UVB组银屑病皮损组织中HSP27、HSP60、HSP90α、HSP90β、Tbet、Foxp3的mRNA表达量以及MCP-1、IFN-γ、IL-17的含量均显著低于对照组,GATA-3、RORγt的mRNA表达量以及IL-10、TGF-β1的含量均显著高于对照组;NB-UVB组患者治疗后银屑病皮损中HSP27、HSP60、HSP90α、HSP90β的mRNA表达量与MCP-1、IFN-γ、IL-17的含量以及T-bet、Foxp3的mRNA表达量呈正相关,与IL-10、TGF-β1的含量以及GATA-3、RORγt的mRNA表达量呈负相关。结论:NB-UVB治疗银屑病能够减少皮损组织中HSPs家族分子的表达量并抑制由HSPs所介导的炎症反应。 Objective:To study the changes of HSPs family expression in psoriasis lesions and their correlation with inflammatory response before and after narrow-band ultraviolet B(NB-UVB)treatment.Methods:Patients who were newly diagnosed with psoriasis vulgaris and treated in Dermatology Department of our hospital between March 2015 and May 2017 were selected and randomly divided into the NB-UVB group who received NB-UVB combined with conventional treatment and the control group who received conventional treatment.Before treatment and 20 days after treatment,proper amount of psoriasis lesion was collected to determine the expression of HSPs family molecules and lymphocyte transcription factors as well as the levels of inflammatory cytokines.Results:Compared with those before treatment,HSP27,HSP60,HSP90α,HSP90β,T-bet and Foxp3 mRNA expression as well as MCP-1,IFN-γand IL-17 levels in psoriasis lesion of both groups significantly decreased whereas GATA-3 and RORγt mRNA expression as well as IL-10 and TGF-β1 levels significantly increased after treatment,and HSP27,HSP60,HSP90α,HSP90β,T-bet and Foxp3 mRNA expression as well as MCP-1,IFN-γand IL-17 levels in psoriasis lesion of NB-UVB group were significantly lower than those of control group whereas GATA-3 and RORγt mRNA expression as well as IL-10 and TGF-β1 levels were significantly higher than those of control group(P 0.05);HSP27,HSP60,HSP90αand HSP90βmRNA expression in psoriasis lesion of NB-UVB group after treatment were positively correlated with MCP-1,IFN-γand IL-17 levels as well as T-bet and Foxp3 mRNA expression,and negatively correlated with IL-10 and TGF-β1 levels as well as GATA-3 and RORγt mRNA expression.Conclusion:NB-UVB treatment of psoriasis can reduce the expression of HSPs family members and inhibit the inflammatory response mediated by HSPs.
