小鼠颞叶联合皮层的突触后电流反应特点

摘要 为研究小鼠颞叶联合皮层(temporal association cortex,TeA)接受信息输入的反应特点,将光遗传技术与离体脑片电生理技术相结合,使用全细胞电压钳记录了C57BL小鼠的TeA锥体神经元的突触后电流反应(EPSC、IPSC),分析其幅度大小、E/I比值、反应延时等指标,得出了不同的反应类型,并统计分析反应类型与神经元细胞膜电生理特性之间的关系。综上得出结论,TeA神经元接受来自听皮层的信息输入有6种反应类型,各类型之间的EPSC和IPSC的幅度有明显差异,说明这些神经元在信息输入过程中起到了不同的作用,并且突触后电流之间的差异与细胞电生理特性无关,这很可能是多突触连接的调节环路和机制导致的结果,对接下来进一步研究TeA信息处理的内部调控环路具有重要意义。 To study the response characteristics of mouse temporal association cortex(TeA)which accepts information input,optogenetic technique was combined with the in vitro electrophysiology to record postsynaptic currents(EPSC,IPSC)on TeA pyramidal neurons of C57 BL mice using whole-cell voltage clamp.To analyze the amplitude,E/Iratio,onset latency and other indicators,the different types of reactions,and statistical analysis of the relationship between the type of reaction and neuronal membrane electrophysiological characteristics were obtained.In conclusion,there are six types of response of TeA neurons in auditory information input,the amplitude of EPSC and IPSC between these types have significant differences,indicating that these neurons play a different role in the process of information input,and the difference of postsynaptic currents is independent of membrane electrophysiological characteristics.It is likely to be the result of a multi-synaptic connection and mechanism,which makes sense for the further study of the internal circuit of TeA information processing.
