黄连素对大鼠心脏缺血再灌注损伤的保护作用 被引量:3

摘要 目的探讨黄连素(berberine,BBR)对大鼠心脏缺血再灌注损伤的作用及其机制。方法将SD大鼠随机均分为假手术组(Sham)、心脏缺血/再灌注损伤组(IRI)和黄连素(低,中,高)组(BBR)(n=10)。通过结扎左冠状动脉前降支构建大鼠MI/R模型。检测各组乳酸脱氢酶(LDH),肌酸激酶同工酶(CK-MB)、天门冬氨酸氨基转移酶(AST)、JAK2、STAT3、p-JAK2、p-STAT3、p-p65、p65、白介素-6(IL-6)、白介素1((IL-1β)和肿瘤坏死因子-α(TNF-α)的表达及心脏病理形态改变。结果与Sham组相比,IRI组CK-MB、AST、LDH、p-JAK2、p-STAT3、p-p65、IL-6、IL-1β和TNF-α的表达以及心脏病理改变均明显增加(P<0.05),而JAK2、STAT3和p65表达无差异(P>0.05)。与IRI组相比,BBR组CK-MB、AST、LDH、p-JAK2、p-STAT3、p-p65、IL-6、IL-1β和TNF-α的表达以及心脏病理改变均明显减少(P<0.05),但JAK2、STAT3和p65表达无差异(P>0.05)。结论黄连素预处理可减轻心脏缺血再灌注损伤,其作用机制与黄连素抑制JAK2/STAT3/NF-KB信号通路激活介导的炎症相关。 Objective To investigate the effect and mechanism of Berberine to myocardial ischemia reperfusion injury in rats.Methods 50 SD rats were randomly equally divided into Sham group(Sham),myocardial ischemia reperfusion injury group(MI/R),and Berberine(low,middle,high)pretreatment group(BBR)(n=10).The model of IRI was induced by ligating left anterior descending coronary artery.The cardiac pathology and the expression of CK-MB,AST,LDH,p-JAK2,p-STAT3,p-p65,IL-6,IL-1,and TNF-in the rats of all groups were examined.Results Compared with Sham group,the IRI group had higher expression of CK-MB,AST,LDH,pJAK2,p-STAT3,p-p65,IL-6,IL-1 and TNF-with higher change of cardiac pathology and there were no statistical significantly differences on the expression of JAK2,STAT3 and p65.Compared with IRI group,the BBR groups had lower expression of CK-MB,AST,LDH,p-JAK2,p-STAT3,p-p65,IL-6,IL-1 and TNF-with lower change of cardiac pathology and there were no statistical significantly differences on the expression of JAK2,STAT3 and p65.Conclusion Berberin can pretreatmentprotect rat against myocardial ischemia reperfusion injury.The mechanism of which is associated with suppressing JAK2/STATA3/NF-KB signal pathway mediating inflammation.
