促红细胞生成素经超声微泡介导治疗大鼠损伤面神经的研究 被引量:1

摘要 目的探讨超声微泡介导促红细胞生成素(EPO)基因修复大鼠受损面神经模型的可行性及有效性。方法选取40只SD大鼠制作面神经损伤模型,随机分为4组,每组10只。A组为EPO+超声组,B组为EPO+微泡组,C组为EPO+超声+微泡组,D组为单纯手术组(PBS)。于EPO基因转染后第1,7,14,28天,观察各组大鼠一般状况,检测损伤面神经传导速度、潜伏期及动作电位波幅。提取损伤处面神经组织,采用RT-PCR检测EPO mRNA的表达。结果转染后第14天,C组大鼠术侧有少量胡须可观察到细微摆动;转染后第28天,C组大鼠一般状况较A组、B组、D组恢复得更好,C组面神经损伤后修复的神经电生理表现优于A组、B组、D组,C组中EPO mRNA表达量明显高于A组、B组、D组(P<0.05)。结论超声微泡介导EPO基因有助于面神经损伤的修复。 Objective To investigate the feasibility and efficacy of erthropoietin( EPO) gene mediated by ultrasound microbubble for treating injured facial nerve of rats models. Methods Totally 40 rats models of facial nerve injury were selected and divided into 4 groups,10 cases in each group. Group A: EPO and ultrasound( EPO + US) group,group B: EPO and microbuble( EPO + MB) group,group C: EPO + ultrasound + microbubble( EPO + MB/US) group, group D: PBS group. On the first, seventh, fourteenth, twenty-eighth day after EPO gene transfection, the general condition of rats in each group was observed, and the nerve conduction velocity( NCV), potential latency and action potential amplitude of injured facial nerve were detected. The injured facial nerve tissue was taken and the expression of EPO mRNA was detected by RT-PCR. Results On the fourteenth day after EPO gene transfection, there was a few beard could be observed in fine wobble on the rats' operated side in group C. On the twenty-eighth day after EPO gene transfection, the general condition of rats in group C recovered better compared with group A,B,D. The nerve electrophysiological expression of injured facial nerve in group C was better than that of group A,B,D. The expression level of EPO mRNA in group C was significantly higher than that of group A,B,D( P〈0. 05). Conclusion EPO gene mediated by ultrasound microbubble is helpful for repairing and regeneration of the injured facial nerve.
