摘要
目的评估汉族人群中子痫前期与多囊卵巢综合征易感基因单核苷酸多态性(包括LHCGR基因rsl3405728位点、THADA基因rsa3429458位点,DENND1A基因rs2479106位点)的遗传相关性。方法选取2014年8月至2016年9月在广东省深圳龙华新区人民医院分娩的汉族187例子痫前期患者和223例正常健康孕妇,分别收集5 ml外周血。纯化外周血DNA后通过PCR扩增LHCGR基因rsl 3405728位点、THADA基因rsa3429458位点,DENND1A基因rs2479106位点,并进行直接测序获得基因型。通过Logistic回归分析研究体质指数(BMI)、年龄以及基因型等因素与子痫前期发病的相关性。结果三类最小等位基因频率在年龄以及体重因素校准后在对照组以及子痫前期组间差异无统计学意义(P>0.05)。具体分析基因型频率时,发现DENND1A SNPrs2479106在病例和对照之间差异有统计学意义(P<0.02,隐性模型),SNP rs13429458在病例与对照组之间差异有统计学意义(P=0.01,累加模型)。结论 LHCGR基因rsl 3405728位点、THADA基因rsa3429458位点,DENND1A基因rs2479106位点多态性与子痫前期具有相关性,可见子痫前期的家族性与多囊卵巢综合征易感基因相关。
Objective To evaluate genetic correlation of preeclampsia and polycystic ovary syndrome susceptibility genes single nucleotide polymorphisms( including LHCGR gene RSL,3405728 THADA loci,THADA gene,rsa3429458 loci,DENND1 A gene rs2479106 loci) of population of Han nationality. Methods 187 cases of patients with preeclampsia and 223 cases of normal pregnant women in hospital delivery from August 2014 to September 2016 were selected as the study objects,and 5 ml peripheral blood of themt were respectively collected. After purification of peripheral blood DNA,LHCGR gene RSL,3405728 locus,THADA gene rsa3429458 locus,and DENND1 A gene rs2479106 locus were amplified by PCR,and genotypes were obtained by direct sequencing. Logistic regression analysis was used to study the relationship between BMI,age,genotype and preeclampsia. Results There were no statistically significant differences of the three type of minimum allele frequency in age and weight factors were adjusted between patients of the control group and patients of the preeclampsia group( P〉0. 05). Genotype frequency was analyzed,it showed that DENND1 A SNP rs2479106 was significantly different between cases and controls( P〈0. 02),and there was significant difference of SNP rs13429458 between the cases and patients of the control group( P = 0. 01,accumulation model). Conclusion There are correlations between LHCGR gene rsl 3405728 locus,THADA gene rsa3429458 locus,DENND1 A geners2479106 locus polymorphism and preeclampsia,and the family preeclampsia is associated with Polycystic ovary syndrome susceptibility gene.
