摘要 利用一种快速、高效的动物肺表面活性物质提取方法,结合自行设计的肺灌洗装置,经超声辅助提取结合高速离心和有机溶剂纯化从猪肺中制得天然肺表面活性物质,个肺提取量为0.412±0.065 g,通过薄层色谱和高效液相色谱等对产物化学成分进行了分析:磷脂种类有6种,含量为89.85±1.09%,蛋白质含量1.06±0.19%,磷脂酰胆碱(PC)含量为61.26%;并由膜天平验证了PS的体外活性,可将表面张力降低到10 m N/m以下。该提纯方法简单、周期短、产率高、毒性低,且不影响PS产物的活性功能,适合于工业化批量生产。 This paper reports a rapid and convenient method for the extraction of pulmonary surfactant(PS)from animal lung. The natural PS was prepared by ultrasonic assisted extraction combined with high-speed centrifugation and organic solvent extraction. The amount of PS extracted from a lung was 0.412 ± 0.065 g,which was determined by thin-layer chromatography and high performance liquid chromatography,the results showed that EPS contained 6 types of phospholipids,phospholipids and protein content of 88.85±1.18% and 1.21±0.13%,respectively,of which phosphatidylcholine content of 61.26%. The in vitro activity of PS was verified by membrane balance,the surface tension was decreased to less than 10 mN/m. The purification method is simple,efficient,low toxicity,and does not affect the activity of PS products,suitable for industrial mass production.
