骨关节炎合并骨质疏松患者血清β-catenin、DKK-1和MMP-9的水平变化及其与临床指标的相关性 被引量:9

摘要 目的探讨膝骨关节炎(KOA)合并骨质疏松(OP)患者血清β-catenin、DKK-1和基质金属蛋白酶-9(MMP-9)的水平及其相关性。方法采用酶联免疫吸附法(ELISA)对60例KOA合并OP患者(OA+OP组),60例KOA患者(OA组),60例OP患者(OP组)及35例体检健康的志愿者(正常组)血清β-catenin、DKK-1和MMP-9水平进行检测,并比较四组受试者血清β-catenin、DKK-1和MMP-9表达水平;分析OA+OP组患者血清β-catenin、DKK-1和MMP-9变化及与红细胞沉降率(ESR)、C-反应蛋白(CRP)、骨密度(BMD)相关性。结果 OA+OP组血清β-catenin水平显著低于正常组和OA组,高于OP组(P<0.05)。OA+OP组血清DKK-1、MMP-9水平显著高于正常组和OA组(P<0.05);OA+OP组血清MMP-9水平高于OP组(P<0.05)。相关性分析结果显示OA+OP组患者血清β-catenin、DKK-1与ESR、CRP呈正相关,MMP-9与ESR、CRP无相关性。β-catenin与腰椎、股骨颈骨密度(BMD)呈正相关,DKK-1、MMP-9与腰椎、股骨颈骨密度(BMD)呈负相关(P<0.05)。结论 OA+OP组患者血清β-catenin、DKK-1、MMP-9表达水平的失调与OA合并OP的发生发展密切相关,与临床指标ESR、CRP及BMD存在一定的相关性。 Objective To investigate the levels of serum β-catenin,DKK-1 and MMP-9 in knee osteoarthritis( KOA) combined with osteoporosis( OP) and their correlation with clinical indexes. Methods ELISA was used to detect the levels of serum β-catenin,DKK-1 and MMP-9 in 60 KOA patients combined with OP( OA + OP group),60 KOA patients( OA group),60 OP patients( OP group) and 35 healthy volunteers( control group). The levels of serum β-catenin,DKK-1 and MMP-9 were compared among the four groups. The correlations of serum β-catenin,DKK-1 and MMP-9 with ESR,CRP and BMD in KOA combined with OP were analyzed.Results The level of serum β-catenin in OA + OP group was significantly lower than that in control group and OA group,but higher than that in OP group( P〈0. 05). The levels of serum DKK-1 and MMP-9 in OA + OP group were significantly higher than those in control group and OA group( P〈0. 05). The level of serum MMP-9 in OA + OP group was higher than that in OP group( P〈0. 05).The serum β-catenin and DKK-1 were positively correlated with ESR and CRP in OA + OP group,while MMP-9 was not correlated with ESR and CRP. β-catenin was positively correlated with lumbar spine and femoral neck BMD,while DKK-1 and MMP-9 were negatively correlated with lumbar spine and femoral neck BMD( P〈0. 05). Conclusion The imbalance of the expression of serum β-catenin,DKK-1 and MMP-9 in OA + OP group is closely related to the occurrence and development of KOA combined with OP,and the expression has a certain correlation with the clinical indicators ESR,CRP and BMD.
