氟对小鼠睾丸间质组织中炎症细胞因子mRNA表达的影响 被引量:1

摘要 [目的]研究氟中毒时睾丸中炎症相关因子的表达情况,探讨其与自身免疫性睾丸炎的相同与不同点。[方法]试验将24只雄性小鼠随机平均分为3组,分别为对照组、NaF组、实验性自身免疫性睾丸炎(EAO)模型组,对照组与EAO组饮用去离子水,NaF组饮用含有100mg·L-1的NaF去离子水,50d后取材,HE染色观察睾丸组织结构的变化,通过qRT-PCR检测睾丸间质组织细胞中炎症相关细胞因子mRNA含量表达量的改变。[结果]试验表明NaF组与EAO组睾丸结构均有不同程度破坏,均出现了生精细胞脱落等情况,在炎症相关因子mRNA的表达情况上,与对照组相比,EAO组IL-6、IL-1β、TNF-α的表达量均显著升高,而IL-23的表达量显著下降,NaF组IL-6、IL-23的表达量显著升高,IL-1β与TNF-α无显著性。[结论]过量氟不仅对睾丸造成了明显的病理学损伤,而且影响了睾丸间质组织炎症因子mRNA的表达,与EAO模型具有一定相似性。 [Objective]The study was designed to detect the effect of fluorosis on the mRNA expression of inflammatory cytokines.The similarities of orchitis h caused by either fluoride or autoimmune system was studied.[Methods]24 male mice were randomly selected and divided into three groups,as control group,NaF group,and experimental autoimmune orchitis(EAO)group.Mice in control and EAO groups were feed with deionized water,and the deionized water containing 100 mg·L-1 NaF supplied for NaF group.Mice were killed and samples were collected after 50 days,and testicle histomorphological changes were observed by HE stain.The mRNA expression variation of inflammatory related cytokines of testicular interstitial cells was detected via qRT-PCR.[Results]Various degrees of damage were observed in mice testicle of both NaF and EAO groups which both showed the exfoliation of spermatogenic cells.Compared to the control group,mRNA expression levels of IL-6,IL-1β,and TNF-αin EAO group were significantly increased,while IL-23 mRNA expression level was significantly decreased in the same group.In mice of NaF group,mRNA expression levels of IL-6 and IL-23 were significantly increased,but IL-1βand TNF-αdidn't show significant change.[Conclusion]Excessive intake of fluoride not only caused obvious pathological damage to the testis,but also affected mRNA expression levels of inflammatory cytokines in testicular tissue of mice,which was similar to the EAO model.
