摘要
猪传染性胃肠炎是由猪传染性胃肠炎病毒(transmissible gastroenteritis virus,TGEV)引起的一种可导致仔猪严重腹泻的高度接触性传染病。本文研究了一株抗TGEV的单链抗体(scFv)对TGEV的动物保护作用。将携带scFv的质粒进行原核表达和纯化,获得了纯化的蛋白。ELISA试验证实scFv与TGEV的特异性反应,空斑试验证实scFv能够抑制病毒的入侵。此外,scFv预先灌服仔猪能够抑制TGEV的感染,给仔猪提供保护。本研究为后续新型抗病毒制剂的研究奠定了基础。
Transmissible gastroenteritis （TGE） is a highly contagious and infectious disease of piglets with diarrhea caused by TGEV（transmissible gastroenteritis virus）. In this study, we test the preventative effect of scFv against TGEV. scFv was purified from bacteriaand identified by SDS-PAGE. The ELISA assay showed the specificity of scFv, and plaque assay confirmed the anti-viral effect ofscFv. Porcine preventing experiment confirmed the scFv could protect porcine from TGEV infection. Our study provided a basis for thefuture development of novel antibody reagent against virus infection.
