荧光定量RT-PCR检测高致病性PRRSV接触感染仔猪增殖动态 被引量:6

摘要 为检测高致病性PRRSV接触感染仔猪的增殖动态,建立了荧光定量RT-PCR方法,该方法在模板为2.18×10~1~2.18×10~6拷贝范围内具有良好线性关系,相关系数为0.996,灵敏度比常规RT-PCR高1 000倍,并且与其他猪病病毒无交叉反应;在3头健康仔猪中引入1头PRRSV人工感染猪,定期监测仔猪接触感染PRRSV后病毒血症和排毒水平,研究结果表明,接触感染猪后分别于3、5 d首次在血清和粪便中检出PRRSV,相比人工接种猪推迟2 d,此后7 d内病毒含量迅速上升并维持较高水平直至死亡。本研究为PRRSV在猪体内的定量检测提供了有效手段,为进一步了解PRRSV的传播机制以及建立防治PRRSV感染的仔猪模型提供了基础数据。
