摘要
目的:研究腺苷激酶(Adenosine kinase,ADK)在肝细胞肝癌(hepatocellular carcinoma,HCC)组织中的表达,并分析其与HCC临床病理特征及预后的关系。方法:采用免疫印迹及免疫组织化学方法检测ADK在HCC组织中的表达情况,并分析其表达和临床病理特征间的相关性。通过Kaplan-Meier法、多因素分析ADK和HCC患者预后的关系。结果:ADK在HCC组织中的表达低于癌旁组织,ADK的表达和组织学分级(P=0.001)、肿瘤大小相关(P=0.024),和其他的临床病理特征无相关性(P>0.05)。Kaplan-Meier生存率曲线表明ADK在HCC中的低表达与患者总生存率的降低显著相关,多因素分析提示ADK是HCC患者一个独立的预后因子。结论:ADK在HCC组织中低表达,是HCC患者一个独立的预后因子,有望成为HCC治疗新的靶点。
Objective To investigate the expression of Adenosine kinase( ADK) in Hepatocellular carcinoma( HCC) and its relationship with clinicopathologic parameters and prognosis of HCC. Methods The expression of ADK in HCC specimens were examined by Western blot analysis and immunohistochemistry,and the correlation between ADK expression and clinicopathological features was analyzed. Kaplan-Meier method and multivariate analysis were used to explore the relationship between ADK and HCC patient's prognosis. Results The expression of ADK in HCC tissues was lower than that in adjacent tissues. The expression of ADK was correlated with histological grade( P = 0.001) and tumor size( P = 0.024),which was not correlated with other clinicopathological features( P〉0.05). The Kaplan-Meier survival curve showed that the low expression of ADK in HCC was significantly associated with the decreased overall survival. Multivariate analysis suggested that ADK was an independent prognostic factor in HCC patients. Conclusion The low expression of ADK in the HCC tissue is an independent prognostic factor for HCC patients and is expected to be a new target for the treatment of HCC.
