摘要
目的:观察加味龟鹿二仙胶汤联合顺铂对小鼠Lewis肺癌组织、骨髓组织TGF-β_1/Smad信号通路的影响。方法:建立气阴两虚型C57BL/6J小鼠Lewis肺癌模型,随机分为正常对照组、模型组、加味龟鹿二仙胶汤组、顺铂组及顺铂+加味龟鹿二仙胶汤低、中、高剂量组,每组20只,干预21 d。观察小鼠瘤质量、抑瘤率及骨髓有核细胞计数,Western Blot检测各组移植瘤组织及骨髓组织中TGF-β_1、Smad4和Smad7的蛋白表达。结果:药物干预后,与模型组比较,联合组肿瘤质量均显著降低,移植瘤组织中TGF-β_1、Smad4表达显著降低,Smad7表达显著升高,骨髓组织中TGF-β_1、Smad4、Smad7表达均显著降低(P<0.01)。加味龟鹿二仙胶汤组骨髓有核细胞计数显著高于模型组(P<0.01)。结论:加味龟鹿二仙胶汤与顺铂联用可显著抑制肿瘤增长,提高抑瘤率,减轻骨髓抑制。其机制可能是通过调节TGF-β_1/Smad信号通路来实现。
Objective:To observe the effect of modified Guilu Erxianjiao decoction combined with cisplatin on TGF-β1/Smad signaling pathway in lewis lung cancer tissues and bone marrow. Methods:Lewis lung cancer model of C57BL/6J mice with deficiency of both vital energy and yin were established and randomly divided into control group, model group, modified Guilu Erxianjiao decoction group, cisplatin group ,cisplatin+modified Guilu Erxianjiao decoction in the low-, medium-and high-dose groups (combined groups ) , with 20 mice in each group. After intervention for 21 d, the tumor mass, tumor inhibition rate and bone marrow nucleated cell count were ob- served. The protein expression of TGF-β1, Smad4 mad Smad7 in each group of transplanted tumor and bone marrow were detected by Western blot. Results:Compared with the model group, the tumor mass decreased significantly( P〈0. 01 ). In the lung cancer tissues ,the protein expression of TGF-β1 and Smad4 were significantly decreased, and the protein expression of Smad7 was significantly increased. In the bone marrow tissue, the protein expression of TGF-β1, Smad4 and Smad7 was significantly decreased(P〈0.01 ). Conclusion:Modified Guilu Erxianjiao decoction combined with cisplatin can significantly inhibit tumor growth,improve tumor inhibition rate, and reduce bone marrow suppression. The mechanism may be achieved by modulating the TGF-β1/Smad signaling pathway.
