有氧运动对2型糖尿病大鼠血管炎症及SIRT1/NF-κB信号通路的影响 被引量:13

Effects of Aerobic Exercise on Vascular Inflammation and SIRT1/NF-κB Signaling Pathway in Type 2 Diabetic Rats

摘要 目的:研究有氧运动对糖尿病大鼠血管炎症的影响,并从SIRT1/NF-κB信号通路探讨其影响机制。方法:采用高脂饲养,小剂量STZ诱导糖尿病大鼠模型。将大鼠分为正常对照组(NC)、糖尿病对照组(DC)和糖尿病运动组(DS)。DS组进行8周游泳训练,1 h/d,5 d/周。8周干预后,采用ELISA、Western blotting和qRT-PCR等方法对血糖代谢、炎症因子和基因表达进行检测。结果:1)DC组大鼠空腹血糖、Glucose AUC、空腹胰岛素均明显高于NC组,胰岛素敏感指数低于NC组;DS组Glucose AUC和空腹胰岛素明显低于DC组。2)DC组大鼠血清TNF-α、IL-1β、MCP-1和VCAM-1均明显高于NC组,DS组均明显低于DC组。3)DC组大鼠SIRT1和Nampt蛋白表达均明显低于NC组,DS组SIRT1和Nampt蛋白及mRNA表达水平均高于DC组;与NC组相比,DC组大鼠NF-κBp65蛋白及mRNA表达显著升高,IκBα蛋白表达降低;与DC组相比,DS组大鼠NF-κBp65蛋白及mRNA表达明显降低,IκBα蛋白表达显著升高。结论:有氧运动能够改善糖尿病大鼠血管炎症,其作用机制可能是通过影响SIRT1/NF-κB信号通路实现的。 Objective: This study aimed at investigating the effect of aerobic exercise on vascular inflammation in diabetic rats and exploring the mechanism through SIRT1/NF-κB signaling pathway. Methods: Animal models for diabetes were established by high-fat diet combined with low-dose STZ injections. Rats were divided into normal control group( NC),diabetes control group( DC) and diabetes swimming group( DS),rats of DS group finished 8 weeks swimming training( 1 h/day,5 days/week). After 8 weeks,the blood glucose metabolism,inflammatory factors and gene expression were measured by ELISA,Western blotting and qRT-PCR. Results: 1) The fasting blood glucose,glucose AUC and fasting insulin in the DC group were significantly higher than those in the NC group,and the insulin sensitivity index was lower than that in the NC group; and the glucose AUC and the fasting insulin in the DS group were significantly lower than those in the group DC. 2) The serum levels of TNF-,IL-1,MCP-1 and VCAM-1 in DC group were significantly higher than those in NC group,while those in DS group were significantly lower than those in DC group. 3) The protein expressions of SIRT1 and Nampt in DC group were significantly lower than those in NC group,and their protein and mRNA expressions in DS group were higher than those in DC group. Compared with group NC,the expression of NF-κBp65 protein and mRNA in DC group increased significantly,and the expression of IκBα decreased; compared with group DC,the expressions of NF-κBp65 protein and mRNA in DS group were significantly decreased,and IκBα protein expression increased significantly. Conclusions: Aerobic exercise can improve vascular inflammation in diabetic rats,and its mechanism may be achieved by affecting the SIRT1/NF-κB signaling pathway.
