胃癌组织标本中lncRNA RN7SK的表达及其筛查价值

Expression of lncRNA RN7SK in tissue samples from gastric cancer patients and its screening diagnostic value

目的检测胃癌组织中长链非编码RNA(lnc RNA)RN7SK表达水平,并探讨其在临床筛查中的价值。方法 q RT-PCR检测lnc RNA RN7SK在胃癌细胞、胃癌组织中的表达水平;绘制ROC曲线,分析其用于筛查胃癌的效能。结果 lnc RNA RN7SK在人胃癌细胞系SGC-7901(3.91±0.53)、MGC-803(3.44±0.29)、HGC-27(4.04±0.87)和BGC-823(4.30±1.13)中的表达水平明显高于人胃粘膜上皮细胞系GES-1(1.02±0.27),差异具有统计学意义(t分别为12.33、7.48、7.20、4.90,P<0.05)。lnc RNA RN7SK在55例胃癌组织中有47例(85.5%)表达上调,8例(14.5%)表达下降。lnc RNA RN7SK筛查胃癌患者的ROC曲线下面积(AUCROC)为0.827,95%可信区间(CI)为0.746~0.907,当cut-off值为0.618时,敏感性为0.836,特异性为0.782。结论 lnc RNA RN7SK在胃癌组织中高表达,且具有较高的筛查效能,可作为潜在的胃癌诊断分子标志物。

Objective To explore the expression of long noncoding RNA（ lnc RNA） RN7 SK and its screening value in clinical diagnosis of gastric cancer. Methods The expression of lnc RNA RN7 SK in gastric cancer cells and paired gastric cancer tissues were measured by q RT-PCR. The screening efficacy was detected by the receiver operating characteristic（ ROC） curve. Results The expression levels of lnc RNA RN7 SK in gastric cancer lines SGC-7901（ 3.91±0.53）,MGC-803（ 3.44± 0.29）,HGC-27（ 4.04± 0.87） and BGC-823（ 4.30±1.13） were markedly higher than that in human gastric mucosal epithelial cell line GES-1,and the difference had statistical significance（ 1.02±0.27,t = 12.33,7.48,7.20 and 4.90,P〈0.05）. The expression of RN7 SK was up-regulated in 85.5%（ 47/55） of gastric cancer tissues while down-regulated in the the other 8 tissues（ 14. 5%）. The area under ROC curve（ AUC^ROC） of lnc RNA RN7 SK expression in gastric cancer patients was 0. 827 and the 95% confidence interval（ CI） was from 0. 746 to 0. 907. When the cut-off value was 0.618,the sensitivity and specificity were 0.836 and 0.782 respectively. Conclusion The level of lnc RNA RN7 SK should be overexpressed in gastric cancer tissues so that it may have high screening efficacy and could be used as a potential molecular marker for the diagnosis of gastric cancer.
