摘要
目的通过基因芯片分析上皮卵巢癌细胞系中差异表达的Lnc RNAs、m RNAs表达谱,为研究上皮卵巢癌相关Lnc RNAs的功能提供实验依据。方法采用基因芯片分析上皮卵巢癌细胞系A2780、HO8910、SKOV3与卵巢上皮细胞系HOSEpi C中Lnc RNAs和m RNAs的表达差异;对差异表达的m RNAs进行KEGG通路富集分析;用q RT-PCR在上皮卵巢癌细胞系及卵巢上皮细胞系中验证差异明显的6条Lnc RNAs的表达水平。结果基因芯片结果发现在3个肿瘤细胞系中上调的Lnc RNAs有227条,下调的有483条;3个上皮卵巢癌细胞系中差异表达的m RNAs主要富集在PI3K-AKT、m TOR及TNF-α等肿瘤相关通路中(P<0.05);PTPRG-AS1、CCNT2-AS1、XLOC_009869、LINC01138在上皮卵巢癌细胞系A2780、SKOV3、和OVCR3中表达上调(P<0.05);RP11-252P19.2、RP11-744I24.2在上皮卵巢癌细胞系A2780、SKOV3、HO8910、OVCR3和3AO中表达下调(P<0.05)。结论基因芯片筛选出在上皮卵巢癌细胞系中差异表达的Lnc RNAs和m RNAs,差异表达m RNAs与PI3K-AKT、m TOR及TNF-α等肿瘤通路有关,有望为卵巢癌的诊断及治疗提供新的靶点。
Objective To Analyze the expression profiles of Lnc RNAs and m RNAs in ovarian epithelial cancer cell lines by gene microarray,and then provide experimental evidences for investigating the function of Lnc RNAs associated with ovarian cancer. Methods The differentially expressed Lnc RNAs and m RNAs in ovarian epithelial cancer cell lines,such as A2780,HO8910 and SKOV3,and ovarian epithelial cell line HOSEpi C were analyzed by gene microarray. The differentially expressed m RNAs were further performed the KEGG pathway enrichment analysis. The expression levels of six candidate Lnc RNAs,which had significant difference between the ovarian epithelial cancer cell line and the ovarian epithelial cell line,were further verified by q RT-PCR. Results There were 227 upregulated Lnc RNAs and 483 down-regulated Lnc RNAs in A2780,HO8910 and SKOV3 cell lines. The differentially expressed m RNAs in A2780,HO8910 and SKOV3 cell lines were mainly enriched in the tumor-related pathways such as PI3 K-AKT,m TOR and TNF-α( P〈0.05). The expression levels of PTPRG-AS1,CCNT2-AS1,XLOC_009869 and LINC01138 in ovarian epithelial cancer A2780,SKOV3 and OVCR3 cell lines were up-regulated( P〈0.05),while those of RP11-252 P19.2 and RP11-744 I24.2 in ovarian epithelial cancer A2780,SKOV3,OVCR3 and 3 AO cell lines were down-regulated( P〈0.05). Conclusion The differentially expressed Lnc RNAs and m RNAs in ovarian epithelial cancer cell lines may be obtained by gene microarray,and the differentially expressed m RNAs are associated with the tumor-related pathways such as PI3 K-AKT,m TOR and TNF-α,which may provide new targets for the diagnosis and treatment of ovarian cancer.
