摘要
通过埃洛石纳米管(HNTs)负载葡萄糖酸氯己定(CG)制备新型的HNTs/CG复合抗菌剂。采用透射电镜表征HNTs的形貌结构,红外光谱表征HNTs/CG复合物的结构,抑菌圈法测试HNTs/CG复合抗菌剂的抗菌性能,扫描电镜观察金黄色葡萄球菌与HNTs/CG复合物作用后形态的变化情况,并通过急性经口毒性实验评价HNTs/CG复合物的短期毒副作用。结果表明:CG药物成功负载在HNTs管腔中,HNTs/CG复合抗菌剂对金黄色葡萄球菌和大肠杆菌均具有良好的抑制效果;金黄色葡萄球菌与HNTs/CG复合物作用后,导致细胞壁破裂而死亡,另外,不同剂量的HNTs/CG复合抗菌剂均未引起小鼠脏器各组织的病理学改变。因此,HNTs/CG复合抗菌剂是一种无毒的新型纳米抗菌复合材料。
An antibacterial drug chlorhexidine gluconate(CG)was introduced to the lumen of halloysite nanotubes(HNTs)to prepare a new antibacterial composite.Transmission electron microscopy(TEM)was used to analyze the morphology of HNTs,the structure of HNTs/CG composite was characterized by fourier transforminfrared(FT-IR),the antibacterial effect was tested by inhibition zone test.The morphology of S.aureus was observed by scanning electron microscopy(SEM)after incubation with HNTs/CG.The acute oral toxicity test was conducted to evaluate the toxic side effects of HNTs/CG in the short term.The results showed that the drug was successfully loaded on HNTs,and the HNTs/CG showed excellent antibacterial activity against Staphylococcus aureus(S.aureus)and Escherichia coli(E.coli).The cell wall of S.aureus incubated with HNTs/CG which disrupted and killed the bacterial.In addition,HNTs/CG did not cause histopathologic changes to the various organs of tested mice.The results suggested that HNTs/CG was a non-toxic new nanoantibacterial composite material.
