摘要
为更加全面的反映维吾尔族药材新塔花的内在质量,对新塔花中的多种成分进行HPLC同步测定,比较不同产地新塔花中咖啡酸、迷迭香酸、蒙花苷的含量。样品制备采用回流提取,选用ZORBAX SB-C18色谱柱(4.6 mm×250 mm,5.0μm);流动相为甲醇(A)-0.2%冰醋酸溶液(B),梯度洗脱,检测波长340 nm,柱温30℃,流速1.0 m L·min-1。咖啡酸、迷迭香酸、蒙花苷分别在240 mg·L-1(r=0.999 9),360 mg·L-1(r=1),7140 mg·L-1(r=0.999 9)呈良好的线性关系,平均回收率分别为100.9%(RS 1.3%),98.25%(RSD 2.0%),101.2%(RSD 1.5%)。测得13批新塔花中咖啡酸质量分数在0.081.07mg·g-1、迷迭香酸质量分数在1.233.59 mg·g-1、蒙花苷质量分数在2.0810.90 mg·g-1。该方法结果准确、重复性好,可用于测定新塔花药材中咖啡酸、迷迭香酸、蒙花苷的含量;3种成分在不同批次的新塔花药材中的含量差异显著,其内在质量差异较为明显。
This study aimed to develop an HPLC method for simultaneous determination of the 3 components in Ziziphora bungeana.The optimum HPLC condition was as follows: ZORBAX SB-C18 column( 4. 6 mm × 250 mm,5. 0 μm) with gradient elution of methanol( A)-0. 2% glacial acetic acid( B),detection wavelength 340 nm,column temperature 30 ℃,flow rate 1 m L·min-1. There were good linearity between peak areas and injection quantity of caffeic acid,rosmarinic acid,and linarin in the range of 2-40 mg·L-1( r =0. 999 9),3-60 mg·L-1( r = 1),7-140 mg·L-1( r = 0. 999 9),respectively. The average recoveries were 100. 9%( RSD 1. 3%),98. 25%( RSD 2. 0%),and 98. 73%( RSD 1. 5%),respectively. The HPLC method was stable and accurate,which could be used to detect caffeic acid,rosmarinic acid,and linarin in Ziziphora bungeana.
