摘要
目的:研究腺苷蛋氨酸对扑热息痛所致急性肝损伤的保护作用。方法:60只雄性昆明小鼠随机分成6组(n=10):正常对照组、模型组、阳性对照组和腺苷蛋氨酸低、中、高剂量(50,100和200 mg·kg^(-1))组。除正常对照组外,采用300 mg·kg^(-1)扑热息痛灌胃造模同时灌胃(ig)给以相应药物,检测血清谷丙转氨酶(ALT)、天冬氨酸转移酶(AST)、肿瘤坏死因子(TNF-α)、白介素1α(IL-1α)水平和肝匀浆中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)含量及小鼠肝脏系数,并用苏木素-伊红(HE)染色处理肝脏组织切片,光镜观察病理学改变,评价腺苷蛋氨酸对扑热息痛所致急性肝损伤的保护作用。结果:腺苷蛋氨酸能够降低扑热息痛所致小鼠急性肝损伤血清ALT,AST和肝脏系数,降低脂质过氧化产物MDA的含量,提高SOD活性,增加GSH储备含量以及抑制炎症因子TNF-α,IL-1α的生成。结论:腺苷蛋氨酸能够预防扑热息痛所致的急性肝损伤。
Objective: To study the protective effects of S-adenosy-L-methionine on acute liver injury induced by acetaminophen in mice. Methods: Sixty KM male mice were randomly divide into six groups( n = 10) :control group,model group,bifendate group,low,medium and high dose S-adenosy-L-methionine( 50,100,and200 mg·kg-1) groups. Excepted for the normal control group,the mice in the other groups were given acetaminophen300 mg·kg-1 by gavage to induce acute liver injury,and corresponding drugs were given. The serum activities of alanine aminotransferase( ALT),aspartate aminotransferase( AST),tumor necrosis factor alpha( TNF-α),interleukin 1 alpha( IL-1α) and the contents of malondialdepyde( MDA),superoxide dismutase( SOD),glutathione( GSH) in the liver tissue were determined,respectively. Liver tissue sections were examined under a light microscope. Objective to evaluate the protective effect of S-adenosy-L-methionine on acute liver injury induced by acetaminophen. Results: In the model of acute liver injury induced by acetaminophen in mice,SAMe could decrease the plasma levels of ALT,AST,and the liver weight,decrease the content of the lipid peroxidation product MDA,increase the reservation of GSH and SOD,and inhibit the generation of inflammtory factors TNF-αand IL-1α. Conclusion: SAMe has protective effect on acute liver injury induced by acetaminophen.
