烟曲霉素发酵条件的初步优化及其扩大培养 被引量:2

摘要 为了获得适宜的烟曲霉素发酵条件,提高烟曲霉素产量。以烟曲霉CEA-1701为生产菌株,运用HPLC测定烟曲霉素含量。通过单因素试验确定各因素范围,进一步运用Plackett-Burman(PB)和Central-Composite Design(CCD)设计优化适合烟曲霉CEA-1701产烟曲霉素的发酵条件,最后对优化的结果进行5L罐的扩大培养。PB试验表明,温度和p H是影响烟曲霉素产量的主要因素;CCD优化得到的产烟曲霉素的发酵条件为:温度32℃、培养基初始p H 6.24、装液量50/250m L、接种量1%(v/v)、玻璃珠添加量5颗。以优化后的条件进行摇瓶和5L发酵罐发酵,烟曲霉素的产量分别提高了18.7%和34.6%,达到73.58mg/L和83.42mg/L。试验优化了烟曲霉CEA-1701产烟曲霉素的发酵条件,初步得到了5L发酵罐的发酵参数,扩大培养显著提高了烟曲霉素的产量。为烟曲霉素的进一步发酵生产提供了依据。 To obtain suitable fumagillin fermentation conditions and improve the production of fumagillin. Aspergillus fumigatus CEA-1701 was used as the producing strain, and the content of fumagillin was determined by HPLC. The range of factors was determined by single factor test. Further, the fermentation conditions of fumagillin produced by Aspergillus fumigatus CEA-1701 were optimized by Plackett-Burman(PB) and Central-Composite Design(CCD). Finally, expending cultivation of 5 L tank was done to the optimized results. PB test showed that temperature and p H were the main indexes affecting the yield of fumagillin. The optimum conditions for producing fumagillin showed by CCD test were as follows: temperature is 32℃, initial p H of the medium is 6.24, liquid volume is 50/250 m L, inoculation amount is 1%(v/v) and beads of glass is 5. Under the optimized conditions,shaked flask and fermented 5 L fermentor. The yield of fumagillin increased by 18.7% and 34.6%, respectively, to73.58 mg/L and 83.42 mg/L. Through the test, the fermentation conditions of fumagillin produced by Aspergillus fumigatus CEA-1701 were optimized, the fermentation parameters of 5 L fermentor were obtained preliminarily,and the growth of fumagillin was improved significantly, which provided a test reference for the further fermentation of fumagillin production.
