摘要
大肠杆菌广泛应用于发酵和代谢工程等生物领域。通过基因工程的手段改造和调控大肠杆菌的代谢途径有着重要的应用价值。近年来,人们利用CRISPR/Cas系统在大肠杆菌中开发了一系列遗传操作工具。本文在简要介绍CRISPR/Cas系统的基础上,重点阐述了其在大肠杆菌基因组编辑和基因表达调控方面的研究现状,结合笔者自身的研究提出开发更高效更精准的基因组编辑工具的思考方向,为在大肠杆菌中应用CRISPR/Cas系统开展相关工作提供参考。
Escherichia coli is widely used in many fields of biological including fermentation and metabolic engineering. In recent years, a series of genetic manipulation tools have been developed in Escherichia coli using the CRISPR/Cas system. In this review, the authors briefly introduced CRISPR/Cas system and then mainly focused on the application of CRISPR/Cas system in genome editing and gene expression regulation of Escherichia coli. Finally,based on our previous research, we envisioned the development of more efficient and precise genome editing tools in the future.
