龙血素A对体外培养人乳腺癌MCF-7细胞增殖、凋亡的影响

Influence of Loureirin A on proliferation and apoptosis of human breast cancer cell MCF-7

观察龙血素A对体外培养人乳腺癌MCF-7细胞增殖、凋亡的影响。采用龙血素A与MCF-7细胞体外共培养(药物组)干预手段,运用四甲基偶氮唑蓝(MTT)法检测细胞的增殖情况;免疫荧光检测细胞增殖标志性蛋白Ki67的表达;免疫印迹检测凋亡调节蛋白Bax、Bcl-XL/S的表达;annexin V-FITC/PI双染流式细胞仪检测细胞的凋亡率。与对照组相比,药物组细胞增殖受到明显抑制,且存在浓度及时间依赖性;药物组Ki67表达显著减少,Bax表达明显增加,Bcl-XL/S表达明显减少;细胞凋亡率增高,上述指标间比较差异均有统计学意义。龙血素A对体外培养的人乳腺癌MCF-7细胞具有抑制增殖和促进凋亡的作用。

To study the effects of Loureirin A on proliferation and apoptosis in human breast cancer cell line MCF-7. Methods： MCF-7 cells were cultured in vitro and treated with Loureirin A. The proliferation of MCF-7 was detected by MTT. Expression of cell proliferation marker protein Ki67 was measured by immunofluorescence. Expression of apoptosis regulating related proteins Bax and Bcl-XL/S was examined by Western blotting. Apoptosis of MCF-7 cells was measured by annexin V-FITC/PI double staining cytometry. Results： Compared with the control group, the experiment groups with Loureirin A treatment presented obviously inhibited proliferation of MCF-7 cells, which was concentration and time dependent. Expression of Ki67 was significantly declined expression of Bax Was dramatically increased and the expression of Bcl-XL/S was notably decreased; apoptosis of cells was prominently increased. Conclusion： Luoreirin A can inhibit proliferation and induce apoptosis of human breast cancer MCF-7 cell.
