摘要
目的观察槲皮素(QUE)对高糖诱导的大鼠系膜细胞氧化应激、单核细胞趋化蛋白1(MCP-1)及细胞外基质表达的影响,探讨QUE治疗糖尿病肾脏疾病(DKD)的理论基础。方法培养大鼠系膜细胞,将系膜细胞分为对照组(Con)、高糖组(HG)和高糖+QUE组(HG+QUE)。采用流式细胞术检测细胞内活性氧簇(ROS)的表达,采用ELISA检测细胞培养上清MCP-1、转化生长因子β1(TGF-β1)和纤连蛋白(FN)的表达。结果培养48h,HG组FN、TGF-β1、MCP-1及ROS表达量均高于HG+QUE组及Con组(P<0.01);与HG组比较,QUE可抑制高糖条件下系膜细胞FN、TGF-β1、MCP-1及ROS的表达(P<0.01)。结论 QUE可通过抑制高糖条件下系膜细胞氧化应激和炎症因子的表达,减少细胞外基质的合成,发挥肾脏保护作用。
Objective To investigate the protective effect of quercetin in mesangial cells induced by high glucose. Methods The cultured rat mesangial cells were divided into control group(Con),high glucose(25 mmol/L)group(HG)and high glucose plus quercetin(50μmol/L)group(HG+QUE).The levels of MCP-1,TGF-β1 and fibronectin(FN)in the supernatant were determined by ELISA.The expression of ROS was detected by flow cytometry. Results At 48 hours of culture,the expression of FN,TGF-β1,MCP-1 and ROS in HG group was higher than that in HG+QUE group and Con group(P〈0.01).QUE inhibited the expression of FN,TGF-β1,MCP-1 and ROS in mesangial cells under high glucose conditions(P〈0.01). Conclusion QUE may play renal protective effects under high glucose conditions by inhibiting oxidative stress,reducing the release of inflammatory factors and reducing the synthesis of extracellular matrix in mesangial cells.
