功能性下颌偏斜大鼠三叉颈核内炎症相关细胞因子和痛觉传递相关神经递质含量变化研究 被引量:1

摘要 目的观察功能性下颌偏斜产生时,三叉颈核内的炎症相关细胞因子和痛觉传递相关神经递质含量的变化,探讨三叉颈核内炎症相关细胞因子和痛觉传递相关神经递质含量与功能性下颌偏斜的相关性,为临床治疗提供理论依据。方法选择7周龄雄性Wistar大鼠,分为空白对照组,实验对照组,功能性下颌偏斜组,功能性下颌偏斜恢复组。又将实验对照组,功能性下颌偏斜组,功能性下颌偏斜恢复组分别分为2周组和4周组。实验大鼠被处死后,取三叉颈核所在部位组织,分别采用ELISA、放免法(radio-immuno-assay,RIA)和液相色谱串联质谱法(liquid chromatography tandem mass spectrometry,LC-MS/MS)对肌肉内的炎症相关细胞因子和痛觉传递相关神经递质的含量进行检测。结果与对照组相比,偏斜组大鼠三叉颈核内炎症相关细胞因子如白介素-6(interleukin-6,IL-6)、白介素-10(interleukin-10,IL-10)和痛觉传递相关神经递质如P物质(substance P,SP)、降钙素基因相关肽(calcitonin gene related peptide,CGRP)、神经肽Y(neuropeptide Y,NPY)及5-羟色胺(5-hydroxytryptamine,5-HT)的含量明显升高,偏斜持续时间越长,升高越明显,待去除偏斜装置下颌位置自然恢复后,与偏斜组比较,以上检测指标在功能恢复组均显示出明显的下降,但与对照组比较,偏斜持续时间越久,这种下降趋势越缓慢。结论三叉颈核内的炎症相关细胞因子和痛觉传递相关神经递质含量变化与下颌偏斜持续时间呈正相关。 Objective To investigate the changes of inflammation-related cytokines and nociceptive transmission associated neurotransmitter levels and its correlation with functional mandible deviation. Methods 7-week-old male Wistar rats were randomly divided into four groups,normal control group,experimental control group,functional mandible deviation group and functional mandible deviation recovery group. The rats were sacrificed,the tissue of the trigeminal neck nucleus was taken and the inflammation-related cytokines and nociceptive transmission associated neurotransmitter levels were examined by enzyme-linked immunosorbent assay, radioimmunoassay andliquid chromatography tandem mass spectrometry. Results The contents of interleukin-6( IL-6),interleukin-10( IL-10),substance P( SP),calcitonin gene related peptide( CGRP),neuropeptide Y( NPY) and 5-hydroxytryptamine( 5-HT) were significantly increased in functional mandible deviation group. The longer the deviation lasted,the more those indices increased. After removing mandibular position deflector,those indices showed significant decrease in functional mandible deviation recovery group. The longer the deviation lasted,the slower it declined in functional mandible deviation recovery group. Conclusion The inflammationrelated cytokines and nociceptive transmission associated neurotransmitter levels were positively correlated with the duration of mandibular deviation.
