摘要
目的研究葛根素对大鼠正畸牙移动后保持阶段牙周组织中成骨标记物Runt相关转录子-2(Runt-related transcription factor-2,Runx2)表达的影响。方法 54只雄性Wistar大鼠,建立正畸牙移动模型,于牙移动最后1天处死6只作为空白组,剩余大鼠随机分为实验组和对照组,局部注射同剂量的葛根素注射液和生理盐水,于保持后的第3、7、14、21d分别处死各组大鼠,上颌骨磨牙段经HE及Runx2免疫组化染色,光镜下观察在保持阶段牙周组织的改建、Runx2的表达以及成骨细胞张力侧数量。结果随着保持时间的延长,实验组与对照组所研究区域出现成骨现象,Runx2表达均升高,在第14d和第21d时,葛根素组Runx2的阳性表达较对照组增加呈差异性(P<0.05)。结论葛根素可以促进牙移动保持阶段牙周组织中Runx2的表达,这可能是葛根素促进大鼠正畸牙移动后牙周组织改建的机制之一。
Objective To investigate the effect of Puerarin on the expression of Runt related transcription factor-2( Runx2) in periodontal tissues of rats after orthodontic tooth movement. Methods A total of 54 male Wistar rats was established as the orthodontic tooth movement model,by the last day of the tooth movement,6 rats were sacrificed as a blank group and the remaining rats were randomly divided into experimental group and control group. The rats were injected with the same dose of normal saline and Puerarin injection. Rats were sacrificed 3,7,14 and 21 days after operation. The maxillary molars were examined by immunohistochemistry and HE. The periodontal tissue remodeling and Runx2 expression were observed at the maintenance stage. Results With the prolongation of maintenance time,osteogenesis and Runx2 expression were increased in the experimental group and the control group. On the fourteenth day and the twenty-first day,the positive expression of Runx2 in puerarin group was different from that in the control group( P〈0. 05). Conclusion Puerarin promoted the expression of Runx2 in periodontal tissue in the retention stage of tooth movemen,which may be one of the mechanisms that puerarin could promote the remodeling of periodontal tissue after orthodontic tooth movement in rats.
