摘要
目的:检测甲状腺旁腺激素样激素(PTHLH)在宫颈癌组织中的表达,并用小干扰RNA(siRNA)沉默宫颈癌HeLa细胞中PTHLH基因的表达,观察PTHLH表达降低对宫颈癌细胞增殖、迁移及侵袭能力的影响,以初步探讨PTHLH在宫颈癌发生中的作用。方法:通过免疫组织化学法检测正常组织与宫颈癌组织中PTHLH蛋白的表达与定位。使用qRT-PCR和免疫荧光对宫颈癌HeLa细胞PTHLH-siRNA组及阴性对照组细胞中PTHLH的表达进行检测。使用CCK-8法检测沉默PTHLH后HeLa细胞与对照组的增殖情况。使用Transwell检测两组细胞迁移及侵袭能力的改变情况。结果:PTHLH在宫颈癌组织中高表达。siRNA能够有效沉默HeLa细胞中PTHLH的mRNA及蛋白的表达。沉默PTHLH表达能够明显抑制HeLa细胞的增殖及侵袭迁移能力。结论:宫颈癌组织中高表达的PTHLH可促进宫颈癌细胞的增殖、侵袭和迁移能力,沉默PTHLH基因可能是控制宫颈癌术后肿瘤复发的有效途径。
Objective:To investigate the expression of parathyroid hormone-like hormone(PTHLH)in the tissues of cervical carcinoma and the effects of PTHLH on HeLa cell proliferation,migration and invasion.Methods:Immunohistochemistry was applied to detect the expression of PTHLH in normal and cervical carcinoma tissues.HeLa cells were divided into two groups,PTHLH-siRNA group(siRNAs were applied to silence the expression of PTHLH in the PTHLH-siRNA group)and negative control group.Quantitative real-time PCR and immunofluorescence were applied to detect the mRNA and protein expression of PTHLH,respectively.CCK-8 and transwell assays were applied to detect the proliferation,migration and invasion ability of HeLa cells in the two groups.Results:PTHLH was highly expressed in the cervical carcinoma tissues while the expression was low in normal tissue.siRNAs effectively silenced the expression of PTHLH with down-regulated mRNA and protein.Furthermore,the proliferation,migration and invasion ability of PTHLH-siRNA group cells decreased significantly compared with the negative control group.Conclusion:PTHLH is highly expressed in the cervical cancer,and might be a key genefor drug target to improve the prognosis.
