摘要 目的探讨解偶联蛋白2(UCP2)在人脑胶质瘤组织标本中的表达及其临床意义。方法收集人脑胶质瘤及正常人脑组织标本,采用免疫组织化学法及Western blot法检测线粒体中UCP2的表达,分析其与临床病理学分级间的关系。结果 UCP2在胶质瘤组织中有阳性表达,阳性率为75.76%;其表达在高级别胶质瘤(HGG)组织中明显高于低级别胶质瘤(LGG),两者差异有统计学意义(P<0.05)。结论 UCP2在人脑胶质瘤组织中有阳性表达,且与肿瘤组织病理分级呈正相关性,表明其在人脑胶质瘤发病中发挥重要作用,有望成为胶质瘤临床治疗的有效靶点之一。 Objective To detect the distribution and expression of uncoupling protein 2( UCP2) and to analyze its relationship with clinicopathological parameters in human glioma. Methods The distribution of UCP2 in human glioma and normal brain tissue was detected by immunohistochemistry. The expression of UCP2 protein in human glioma and normal brain tissue was detected by Western blot. The relationship between UCP2 expression and clinicopathological parameters in glioma was then analyzed. Results UCP2 was overexpressed in human glioma tissue,but was almost undetectable in normal brain tissue. The positive rate of UCP2 expression in human glioma tissue were 75. 76%. Western blot analyses showed that the expression levels of UCP2 protein in glioma tissue were higher than those in normal brain tissue. The expression levels of UCP2 in patients with high grade glioma( HGG) were significantly higher than that in patients with low grade glioma( LGG)( P〈0. 05). Conclusion The expression level of UCP2 is higher in human glioma tissue than in normal brain tissue and it is positively correlated with the pathological grade. It suggests that UCP2 plays an important role in the etiopathogenesis of glioma,so it could be a promising molecular target of gene therapy for human glioma.
