MLH1、MSH6表达与结直肠癌临床病理特征的关系

The relationship between the expression of MLH1 and MSH6 and clinicopathological features of colorectal cancer

目的通过研究错配修复基因MSH6、MLH1在结直肠癌中的表达情况,分析其与结直肠癌临床病理特征的关系,并探讨其作为结直肠癌肿瘤标记物的可行性。方法回顾性分析255例结直肠癌患者的临床及病理资料,以及同期行电子结肠镜检查的30例结直肠黏膜正常者、30例结直肠腺瘤患者的相关资料。分别比较错配修复基因MLH1、MSH6在结直肠癌、结直肠腺瘤以及正常结直肠组织中的表达是否存在差异,分析MLH1、MSH6表达与结直肠癌临床病理特征的关系。结果结直肠癌中MLH1、MSH6蛋白的表达缺失率均明显高于结直肠腺瘤及正常结直肠组织(P<0.05)。不同肿瘤部位、不同淋巴结转移情况、不同TNM分期的结直肠癌中MLH1蛋白的表达缺失程度不同(P<0.05);不同年龄、不同淋巴结转移情况、不同TNM分期的结直肠癌中MSH6蛋白的表达缺失程度不同(P<0.05)。结论结直肠癌中MLH1、MSH6的表达缺失率明显高于结直肠腺瘤及正常结直肠组织,MLH1、MSH6在不同临床病理特征的结直肠癌中的表达存在显著差异,MLH1、MSH6可能作为结直肠癌诊断的标志物。

Objective Through the study of mismatch repair genes MSH6 and MLH1 expression in colorectal cancer,the relationship between the genes and clinicopathological features of colorectal cancer was analyzed. and investigate if MSH6 and MLH1 can as tumor markers for colorectal cancer. Methods Retrospective analysis of clinical and pathological data of 255 cases of colorectal cancer and related data of 30 cases of normal colorectal mucosa and 30 cases of colorectal. The expression of mismatch repair genes MSH6 and MLH1 in colorectal carcinoma,colorectal adenoma and normal colorectal tissues were compared; Analysis of the relationship between MSH6 and MLH1 and clinical pathological characteristics of colorectal cancer. Results Expression of MLH1 and MSH6 protein in colorectal carcinoma was significantly higher than that in colorectal adenoma and normal colorectal tissue,and the difference was statistically significant（ P 〈0. 05）. The expression of MLH1 protein was different in different tumor sites,lymph node metastasis and TNM stages,and the difference was statistically significant（ P 〈0. 05）. The expression of MSH6 protein was different in different ages,different lymph node metastasis and different TNM stage,and the difference was statistically significant（ P〈 0. 05）. Conclusion The rate of deletion of MLH1 and MSH6 in CRC was significantly higher than that in colorectal adenoma and normal colorectal tissue. The expression of MLH1 and MSH6 in colorectal carcinoma are significantly correlated. MLH1 and MSH6 and may serve as a marker for the diagnosis of colorectal cancer.
