蒙古沙冬青AmNAC1和AmNAC2的表达分析与植物表达载体构建 被引量:4

Expression Analysis and Plant Expression Vector Construction of AmNAC1 and AmNAC2 from Ammopiptanthus mongolicus

摘要 NAC转录因子为植物所特有,是植物抗逆信号转导途径的关键组分,可通过激活许多抗逆功能基因的表达而使植物形成抗逆保护机制。本研究利用RT-qPCR方法分析了强抗逆植物蒙古沙冬青AmNAC1和AmNAC2基因在低温和脱水胁迫下的表达变化。结果表明其表达均受这两种胁迫的诱导而显著上调。同时利用PCR方法克隆到这两个基因的编码区cDNA和gDNA。其gDNA中均含有两个内含子序列。推测AmNAC1和AmNAC2的编码蛋白分别由351个和292个氨基酸残基组成,其氨基酸序列中均含有一个保守的NAC结构域。此外成功构建了两个基因的植物超表达载体。这些工作为进一步鉴定AmNAC1和AmNAC2的功能提供了参考依据。 NAC transcription factors are unique to plants and are key components in the anti-signal transduction pathways of plants. They can make plants to form protective mechanisms against stresses by activating the expressions of many stress-tolerant genes. In this research, RT-q PCR method was applied to analyze the expression changes of AmNAC1 and AmNAC2 genes in Ammopiptanthus mongolicus under cold and dehydration treatments which is a plant with very strong resistance to adverse environments. The results showed that the expressions of both the two genes were significantly induced by these stresses. At the same time, we also cloned the expressions into the coding region c DNA and g DNA of the two genes using PCR method. Both of the two g DNA contained two introns sequences. The encoded protein of AmNAC1 and AmNAC2 were predicted to be composed of 351 and292 amino acid residues respectively, and both of their amino acid sequences contained a conserved NAC domain.Moreover, we successfully constructed the plant over-expression vectors of AmNAC1 and AmNAC2. This work might provide reference for further functional identification of AmNAC1 and AmNAC2 in the future.
