西部沙樱枝条愈伤组织诱导、增殖及分化培养 被引量:1

Induction, Multiplication and Differentiation Culture of Callus in Branches of Prunus besseyi

摘要 以西部沙樱一年生枝条为材料,研究不同激素配比条件下外植体愈伤组织的诱导、增殖及分化诱导。结果表明:西部沙樱茎段在2,4-D 0.5 mg/L+6-BA 0.5 mg/L+3%蔗糖+0.6%琼脂(pH 5.75)的MS培养基上可诱导出愈伤组织;愈伤组织增殖的最佳培养基为MS+2,4-D 0.5 mg/L+6-BA 1.0 mg/L+3%蔗糖+0.6%琼脂(pH 5.75);愈伤组织分化的最佳培养基为MS+NAA 0.3 mg/L+6-BA 1.0 mg/L+3%蔗糖+0.6%琼脂(pH 5.75),经分化可获得无性系芽,继而产生丛生苗。西部沙樱愈伤诱导及分化的成功为西部沙樱资源的保护、开发和利用提供了理论依据和技术保障。 In this research, one-year-old branches of Prunus besseyi were selected as the material to study induction,proliferation and differentiation of callus in explants under different hormone ratio. The results showed that stem segments of Prunus besseyi could induce callus in MS medium of 2,4-D 0.5 mg/L+6-BA 0.5 mg/L+3% sucrose and 0.6% agar(pH 5.75). The best medium for callus proliferation was MS+2,4-D 0.5 mg/L+6-BA 1.0 mg/L+3%sucrose+0.6% agar at pH 5.75. The best medium for callus differentiation was MS+NAA 0.3 mg/L+6-BA 1.0 mg/L+3% sucrose +0.6% agarose at pH 5.75. Clonal buds could be obtained after differentiation, and then clumping seedlings were produced. The success of callus induction and differentiation of Prunus besseyi could provide theoretical basis and technical support for the conservation, development and utilization of Prunus besseyi resources.
