摘要
目的研究鱼藤酮诱导的帕金森病小鼠模型血压及焦虑抑郁行为的变化。方法 12月龄C57BL/6雄性小鼠360只,随机分为两组,各180只。实验组给予鱼藤酮5mg/(kg·d)灌胃,对照组给予等量不含鱼藤酮的溶剂。两组分别于给药1周、2周、3周、1月、2月、3月后进行鼠尾血压测量及旷场检测。结果两组各时间点平均动脉压比较,差异无显著性(P>0.05)。旷场检测结果显示,实验组给药2周周围区域活动时间与中央区域活动时间的比值较对照组明显增大,差异有统计学意义(t=3.019,P<0.01)。结论鱼藤酮灌胃给药不能使C57BL/6小鼠的平均动脉压发生变化,鱼藤酮短时间给药能够使小鼠产生焦虑样行为。
Objective To examine the changes in blood pressure and anxiety in mice with rotenone-induced Parkinson disease(PD). Methods A total of 360 male C57BL/6 mice at 12 months of age were randomly divided into control group and experimental group(n=180 per group).The experimental group was gavaged with 5 mg/(kg·d)rotenone(RN),and the control group was gavaged with the same volume of solvent vehicle.Blood pressure measurement and Open Field Test(OFT)were performed at 1 week,2 weeks,3 weeks,1 month,2 months,and 3 months post-treatment. Results There was no significant difference in mean arterial pressure(MAP)at each post-treatment time point between the two groups(P〉0.05).The ratio of peripheral time to central time of OFT at 2 weeks post-treatment was significantly increased in the experimental group than in control group,the difference is significant(t=3.019,P〈0.01). Conclusion Oral gavage of RN does not result in MAP change,but can induce anxiety in C57BL/6 mice.
