动脉球囊损伤后主动脉组织中Atox1和Lox表达及缬沙坦对其影响

EFFECT OF VALSARTAN ON THE EXPRESSION OF ATOX1 AND LOX AFTER ARTERY BALLOON INJURY

摘要 目的研究血管内皮球囊损伤后铜伴侣蛋白-1(Atox1)、赖氨酸氧化酶(Lox)的表达及缬沙坦对其影响,探讨Atox1、Lox及缬沙坦在血管再狭窄中的作用。方法 Wistar大鼠36只,随机分为对照组、手术组和药物组,各12只。制备大鼠主动脉球囊损伤后再狭窄动物模型,药物组给予缬沙坦灌胃,对照组、手术组给予生理盐水灌胃,各组分别于术后14、28d取6只大鼠腹主动脉,检测主动脉内膜、中膜厚度,Atox1、Lox mRNA及蛋白表达。结果与对照组比较,手术组术后14、28d主动脉内膜、中膜增厚,Atox1、Lox mRNA及蛋白表达明显上升;与手术组比较,药物组术后14、28d主动脉内膜、中膜增生减轻,Atox1、Lox mRNA及蛋白表达下降,差异有显著性(F=25.73~34.16,P<0.05)。结论 Atox1、Lox参与大鼠主动脉球囊损伤后内膜增生过程;缬沙坦可抑制内膜增生,其作用与下调Atox1、Lox的表达有关。 Objective To investigate the expression of the copper chaperone Atox1 and lysyl oxidase(Lox)after vascular endothelial balloon injury,the effect of valsartan,and the role of Atox1,Lox,and valsartan in vascular restenosis. Methods A total of 36 Wistar rats were randomly divided into control group,surgery group,and drug group,with 12 rats in each group.A rat model of restenosis after aortic balloon injury was established.The rats in the drug group were given valsartan by gavage,and those in the control group and the surgery group were given normal saline by gavage.The abdominal aorta was harvested from 6 rats on days 14 and 28 after surgery to measure aortic intima-media thickness,and the mRNA and protein expression of Atox1 and Lox was also measured. Results Compared with the control group,the surgery group had significant increases in aortic intima-media thickness and the mRNA and protein expression of Atox1 and Lox on days 14 and 28 after surgery;compared with the surgery group,the drug group had significant alleviation of aortic intima-media hyperplasia and significant reductions in the mRNA and protein expression of Atox1 and Lox on days 14 and 28 after surgery(F=25.73-34.16,P〈0.05). Conclusion Atox1 and Lox participate in the process of intimal hyperplasia after aortic balloon injury in rats,and valsartan can inhibit intimal hyperplasia by downregulating the expression of Atox1 and Lox.
