小鼠α-syn基因敲除对高铁所致黑质酪氨酸羟化酶表达降低影响 被引量:2

EFFECT OF α-SYNUCLEIN GENE KNOCKOUT ON REDUCED EXPRESSION OF TYROSINE HYDROXYLASE IN THE SUBSTANTIA NIGRA INDUCED BY A HIGH IRON LEVEL

摘要 目的探讨α-突触核蛋白(α-syn)基因敲除(α-syn^(-/-))小鼠是否能够抵抗高铁饮食造成的黑质(SN)酪氨酸羟化酶(TH)表达降低。方法实验分为野生型正常饮食组、野生型高铁饮食组、α-syn^(-/-)正常饮食组和α-syn^(-/-)高铁饮食组。其中高铁饮食组用含质量分数0.03羰基铁粉的鼠粮喂养小鼠3个月。采用蛋白质免疫印迹实验检测各组小鼠TH的表达。结果与野生型正常饮食组相比,野生型高铁饮食组小鼠TH蛋白表达水平降低(F=3.761,P<0.05);α-syn^(-/-)高铁饮食组和α-syn^(-/-)正常饮食组小鼠TH蛋白表达水平比较差异无显著性(P>0.05)。结论α-syn^(-/-)小鼠能够抵抗高铁负载造成的SN区TH表达下降,减轻铁对SN神经元的损伤。 Objective To investigate whetherα-synuclein(α-syn)-knockout(α-syn^-/-)mice can resist the reduced expression of tyrosine hydroxylase(TH)in the substantia nigra(SN)caused by a high iron level. Methods The mice were divided into wild-type+normal diet group,wild-type+high-iron diet group,α-syn^-/-+normal diet group,andα-syn^-/-+high-iron diet group.The mice in the wild-type+high-iron diet group and theα-syn^-/-+high-iron diet group were given the diet with a mass fraction of carbonyl iron powder of 0.03 for 3 months.Western blotting was used to measure the expression of TH in each group.Results Compared with the wild-type+normal diet group,the wild-type+high-iron diet group had a significant reduction in the protein expression of TH(F=3.761,P〈0.05).There was no significant difference in the protein expression of TH between theα-syn^-/-+high-iron diet group and theα-syn^-/-+normal diet group(P〉0.05). Conclusion Theα-syn^-/-mice can resist the reduction in TH in the SN induced by iron and alleviate the damage of SN neurons caused by iron.
